去氢骆驼蓬碱抗骨肉瘤作用机制研究

摘要 目的探讨去氢骆驼蓬碱抗骨肉瘤的作用机制。方法本实验分为体外实验和体内实验。体外实验中,设置不同浓度的去氢骆驼蓬碱组及空白对照组,测定去氢骆驼蓬碱对处于生长期的人骨肉瘤细胞的影响:借助细胞活力测定(MTS)法对细胞增殖的影响进行探讨;通过划痕实验分析去氢骆驼蓬碱的侵袭能力,通过Transwell实验分析去氢骆驼蓬碱的迁移能力;应用流式细胞仪(FCM)分析去氢骆驼蓬碱对骨肉瘤细胞周期的影响,并观察其中的诱导凋亡效应及相关分子机制。体内实验开展裸鼠荷瘤实验,实验完毕后,比较肿瘤的重量、裸鼠的重量,并进行免疫组化切片对肝、肺、脾进行HE染色,观察其中的抗肿瘤效果。结果体外实验表明,去氢骆驼蓬碱对细胞增殖具有一定的抑制效果(P<0.05),且去氢骆驼蓬碱组的抗凋亡基因相对表达量、相对迁移能力以及侵袭能力明显低于空白对照组(P<0.05)。FCM分析显示,随着去氢骆驼蓬碱浓度的增加,凋亡率也明显增加(P<0.05)。体内实验表明,去氢骆驼蓬碱能够明显抑制Saos-2移植瘤。结论去氢骆驼蓬碱对骨肉瘤细胞的增殖、迁移、侵袭有明显的抑制作用,具有较高的应用价值,值得临床推广。 Objective To explore mechanism of action of Harmine(hm)for osteosarcoma and provide new ideas for clinical treatment.Methods The experiment included in vitro and in vivo parts.Different concentrations of dehydroalkaline and blank control group were established for human osteosarcoma cells at growth stage for vitro experiment to explore effect on cell proliferation with MTS method.Invasive ability of dehydroalkaline was analyzed through scratch test,migration ability of dehydroalkaline was analyzed through Transwell test,effect of dehydroalkaline on cell cycle of osteosarcoma cells was explored with FCM,and inducing effect of apoptosis and related molecular mechanisms was observed.During in vivo experiment,weight of tumor and nude mouse was compared after nude mouse tumor bearing experiment,immunohistochemical sections were carried out to stain liver,lungs,and spleen with HE to observe anti-tumor effect.Results In vitro experiments showed harmine had a certain inhibitory effect on cell proliferation,relative expression level,migration ability,and invasion ability of anti apoptotic genes was significantly lower than blank control group,indicating harmine had a certain inhibitory effect on the proliferation,migration,and invasion of osteosarcoma cells.According to FCM analysis,based on different concentrations,apoptosis rate significantly increased with increase of concentration(P<0.05).Vivo experiments showed dehydroalkaline could significantly inhibit Saos-2 transplanted tumors.Conclusion Dehydroharmine(hm)can achieve significant inhibitory effect and high application value on osteosarcoma,which is worthy of clinical promotion.
