空心多孔金纳米酶的制备及其对Hg^(2+)和Ag^+的比色检测

摘要 开发基于纳米酶的重金属离子比色传感分析方法,进而克服纳米酶在近中性p H环境催化活性较低的不足,仍然是当前面临的一项重要挑战.文中通过一步法制备了空心多孔金纳米酶(HPAuNSs),采用TEM,XRD,EDS,XPS等对纳米酶进行了表征,并将HPAuNSs用于Hg^(2+)和Ag^(+)的比色分析检测.结果表明,Hg^(2+)和Ag^(+)的加入均显著增强HPAuNSs在近中性条件下的类过氧化物酶(POX)活性,提高了H_(2)O_(2)的分解速率并加速底物3,3',5,5'-四甲基联苯胺(TMB)的氧化反应.利用显色底物TMB氧化后蓝色溶液的颜色变化,成功实现了Hg^(2+)和Ag^(+)的特异性分析检测,检测限分别为7.43nmol·L^(-1)和0.89μmol·L^(-1).构建的纳米酶传感体系打破了纳米酶活性的p H限制,以裸眼可见的溶液体系颜色变化为读出信号,实现了两种重金属离子的检测,为拓宽纳米酶在分析传感领域的应用提供了参考. The development of nanozyme-based colorimetric sensing methods for heavy metal ions,especially to overcome the limitation of the low catalytic activity of nanozymes in near-neutral pH environments,is still an important challenge.In this study,hollow porous gold nanozymes(HP Au NSs)were prepared by a one-step method,and successfully characterized by TEM,XRD,EDS and XPS.The HP Au NSs were used for colorimetric detection of Hg^(2+)and Ag^(+) ions.The results showed that the addition of Hg^(2+)and Ag^(+) significantly can enhances the peroxidase-like(POX)activity of the HP Au NSs under near-neutral conditions,increase the decomposition rate of H_(2)O_(2),and accelerate the oxidation reaction of the substrate 3,3′,5,5′-tetramethylbenzidine(TMB).The specific analysis and detection of Hg^(2+)and Ag^(+) are successfully achieved by using the colour change of the blue solution after oxidation of the chromogenic substrate TMB,and the detection limits are 7.43 nmol·L^(-1)and 0.89μmol·L^(-1),respectively.The constructed nanozyme sensing system breaks the pH limitation of nanozyme activity,and uses the colour change of the solution system visible to the naked eye as the readout signal to realize the detection of two heavy metal ion pollutants,which provides a reference for broadening the application of nanozyme in the field of analytical sensing.
