摘要
目的观察针刺对阿尔茨海默病(Alzheimer′s disease,AD)模型快速老化小鼠(senescence-accelerated mouse-prone 8,SAMP8)学习记忆能力及淋巴细胞介导的免疫炎症的影响,探讨针刺治疗AD的部分作用机制。方法将12只6月龄的雄性SAMP8小鼠随机分为模型组和针刺组(各6只),另取6只同月龄雄性SAMR1小鼠作为空白组。针刺组小鼠选取“百会”和“涌泉”进行40 d的针刺干预。治疗结束后,采用水迷宫实验检测各组小鼠的学习和记忆能力,流式细胞技术检测小鼠脾脏中淋巴细胞的细胞亚群CD_(4)^(+)T、CD_(8)^(+)T比例和免疫分子CD3、CD22的表达量,酶联免疫吸附试验检测大脑皮层中IL-1β、IL-6和TNF-α的表达以及血清中IL-2和IFN-γ的表达水平。结果与空白组比较,模型组小鼠逃避潜伏期延长(P<0.05)、穿越平台次数明显减少(P<0.01)、CD_(4)^(+)T比例和CD22表达量降低(P<0.05)、CD_(8)^(+)T比例和CD3表达量降低(P<0.01)、CD_(4)^(+)T/CD_(8)^(+)T比值升高(P<0.01)、IL-1β、IL-6和TNF-α含量升高(P<0.01)、IL-2和IFN-γ含量明显降低(P<0.01);与模型组比较,针刺组小鼠第1、2、3天的平均逃避潜伏期明显缩短(P<0.01)、穿越平台次数增加(P<0.05)、CD_(4)^(+)T和CD_(8)^(+)T比例升高(P<0.05)、IL-6和TNF-α水平下降(P<0.01,P<0.05)、IL-2和IFN-γ水平升高(P<0.05)。结论针刺“百会”“涌泉”对SAMP8小鼠空间学习和记忆能力的提升,可能与改善淋巴细胞介导的免疫炎症反应有关。
Objective To observe the effects of acupuncture on the learning and memory abilities and lymphocyte-mediated immune inflammation in senescence-accelerated mouse-prone 8(SAMP8),a model of Alzheimer′s disease(AD),and to explore some of the mechanisms of action of acupuncture in the treatment of AD.Methods Twelve 6-month-old male SAMP8 mice were randomly divided into model group and acupuncture group(6 mice in each),and 6 same-month-old male SAMR1 mice were taken as the blank group.The mice in the acupuncture group underwent acupuncture intervention at“Baihui(DU20)”and“Yongquan(KI1)”acupoints for 40 days.After the acupuncture treatment,the learning and memory abilities of mice in each group were detected by Morris Water Maze test,the ratio of cell subsets CD_(4)^(+)T and CD_(8)^(+)T and the expressions of immune molecules CD3 and CD22 in lymphocytes in the spleen of mice were detected by flow cytometry,and the expressions of IL-1β,IL-6 and TNF-αin the cerebral cortex and the expression levels of IL-2 and IFN-γin serum were detected by enzyme-linked immunosorbent assay.Results Compared with the blank group,mice in the model group had prolonged escape latency(P<0.05),decreased number of crossing the platform(P<0.01),reduced CD_(4)^(+)T ratio and CD22 expression(P<0.05),reduced CD_(8)^(+)T ratio and CD3 expression(P<0.01),and increased CD_(4)^(+)T/CD_(8)^(+)T ratio(P<0.01).Besides,the contents of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were increased in SAMP8 mice(P<0.01),while the contents of interleukin-2(IL-2)and interferon-γ(IFN-γ)were decreased(P<0.01).Compared with those in the model group,the mice in group A demonstrated a shorter escape latency from the 1st day to the 4th day(P<0.01),and the number of crossing the platform was increased(P<0.05).Moreover,the proportion of CD_(4)^(+)T and CD_(8)^(+)T was increased(P<0.05).The levels of IL-6 and TNF-αwere significantly decreased(P<0.01,P<0.05)and the levels of IL-2 and IFN-γwere increased(P<0.05).Conclusion Acupuncture at“Baihui(DU20)”and“Yongquan(KI1)”acupoints can improve the spatial learning and memory abilities of SAMP8 mice,which may be related to the improvement of lymphocyt-mediated immune inflammatory response.
