摘要
目的:探讨小鼠接受骨髓移植8周后肥大细胞和嗜碱性粒细胞的重建,并确定其分化来源。方法:向接受致死照射剂量的C57BL/6受体小鼠(CD45.2)尾静脉注射同种同基因小鼠(CD45.1)全骨髓单个核细胞(wBMMNC)5×10^(6)个。移植后第8周取对照组(未经照射的CD45.2小鼠)和移植组小鼠外周血、腹腔灌洗液、脾脏和骨髓细胞,制备细胞悬液并裂解红细胞,计数有核细胞总数,并利用流式抗体进行染色,分析不同组织中肥大细胞及嗜碱性粒细胞的比例和细胞数,并通过对细胞中CD45.1和CD45.2的占比情况分析其供受体分化来源。结果:小鼠骨髓移植8周后,腹膜腔中肥大细胞比例和骨髓中嗜碱性粒细胞比例均恢复正常,与对照组无显著差异;外周血和脾脏中嗜碱性粒细胞比例也有所恢复,与对照组有显著差异(P<0.05)。腹膜腔肥大细胞中CD45.2阳性细胞占比约为93.4%,外周血、脾脏以及骨髓细胞嗜碱性粒细胞中CD45.1阳性细胞占比分别为87.6%、96.4%、90.8%。结论:小鼠经骨髓移植8周后,腹膜腔中肥大细胞和骨髓中嗜碱性粒细胞重建较快,外周血和脾脏中嗜碱性粒细胞重建相对较慢。同时,肥大细胞分化来源于宿主自体驻留细胞,而嗜碱性粒细胞分化则来源于供体骨髓细胞。
Objective:To investigate the reconstitution and origin of mast cells and basophils in mice after bone marrow trans-plantation for 8 weeks.Methods:Total 5×10^(6) whole bone marrow mononuclear cells(wBMMNC)from C57BL/6 homogeneic mice(CD45.1)were injected into the tail vein of recipient mice(CD45.2)which were received lethal irradiation.Eight weeks after trans-plantation,peripheral blood,peritoneal lavage fluid,spleen and bone marrow of control and transplantation groups were collected.Cell suspension were prepared and red blood cells were lysed,and the total number of nucleated cells were counted.Flow cytometry antibodies were used for staining,and the proportion and number of mast cells and basophils in different tissues were analyzed.Simul-taneously,the cell origin was identified by analyzing the proportion of CD45.1 and CD45.2.Results:After 8 weeks of bone marrow transplantation,the proportion of mast cells in peritoneal cavity and basophils in bone marrow returned to the normal level,and there was no significant with control.The proportions of basophils in peripheral blood and spleen were also reconstituted,but it was different with control group(P<0.05).The percentage of CD45.2+mast cells in peritoneal was about 93.4%,and the percentage of CD45.1+ba-sophils cells in peripheral blood,spleen and bone marrow were 87.6%,96.4%and 90.8%,respectively.Conclusion:After 8 weeks of bone marrow transplantation,mast cells in peritoneal cavity and basophils in bone marrow have been reconstituted quickly,while basophils in peripheral blood and spleen reconstituted relatively slowly.Meanwhile,mast cells differentiate from host resident cells and basophils differentiate from donor bone marrow cells.
