高富硒酵母菌的筛选鉴定及其产香气物质成分分析

摘要 结合耐硒法和红硒法从安康市平利县富硒土壤和建始县关口葡萄样品中筛选具有高富硒能力的酵母菌,通过形态学观察、生理生化试验和分子生物学技术对菌株进行鉴定,同时对其生长特性进行研究,并利用顶空固相微萃取-气相色谱-质谱联用(HS-SPME-GC-MS)技术对该菌株富硒发酵液中的香气成分进行分析。结果表明,从土壤和葡萄样品中筛选鉴定出一株具有高富硒能力的戴尔有孢圆酵母(Torulaspora delbrueckii)G-1,其在含有50 mg/L Na2SeO3的YPD液体培养基中培养36 h后,硒含量为(1.64±0.034)mg/g,生物量为(4.80±0.034)g/L。该菌株可耐受葡萄糖200 g/L、SO2300 mg/L,乙醇体积分数6%。从戴尔有孢圆酵母G-1发酵液中共检出33种挥发性香气物质,包括醇类8种、酯类10种、酸类3种、醛酮类5种、烷烃类3种以及其他类4种。与未加硒发酵液相比,富硒发酵液中增加了17种独特的挥发性化合物(如苯乙醇、9-十六碳烯酸乙酯、壬酸等),丰富了发酵液的花香、酯香和蜡香。 Combined with selenium tolerance method and red selenium method,yeasts with high selenium enrichment ability were screened from selenium-enriched soil in Pingli county of Ankang city and Guankou grape samples in Jianshi county.The strains were identified by morphological observation,physiological and biochemical tests and molecular biology techniques.The growth characteristics were studied,and the aroma components in the selenium-enriched fermentation broth of the strain were analyzed by headspace solid phase microextraction-gas chromatography-mass spectrometry(HS-SPME-GC-MS).The results showed that a strain of Torulaspora delbrueckii G-1 with high selenium enrichment ability was screened and identified from soil and grape samples.After being cultured in YPD liquid medium containing Na2SeO350 mg/L for 36 h,the selenium contents and biomass were(1.64±0.034)mg/g and(4.80±0.034)g/L,respectively.The strain G-1 could tolerate glucose 200 g/L,SO2300 mg/L and ethanol 6%(V/V).A total of 33 volatile aroma compounds were detected from the fermentation broth of T.delbrueckii G-1,including 8 alcohols,10 esters,3 acids,5 aldehydes and ketones,3 alkanes and 4 others.Compared with the selenium-free fermentation broth,17 unique volatile compounds(such as phenylethyl alcohol,ethyl 9-hexadecenoate,nonanoic acid,etc.)were added in the selenium-enriched fermentation broth,which enriched the floral,ester and wax aroma of the fermentation broth.
