龙岩地方品种鸡精液ALV-J检测与gp85基因分析

Molecular epidemiology and analysis of avian leukosis virus subgroup J(ALV-J)in indigenous chickens from the Longyan region

摘要 为掌握龙岩地方品种鸡精液中的J亚型禽白血病病毒(ALV-J)感染率与流行毒株的分子特性,对龙岩地方品种鸡128份精液样本进行p27抗原ELISA检测及ALV-J gp85基因PCR检测。结果表明,17.2%的样本呈现p27抗原阳性,而gp85基因PCR检测确认5.5%的样本含有ALV-J。进一步基因序列分析,发现这些毒株与J亚群参考毒株间的高度同源性,核苷酸序列相似度为92.0%~97.9%,特别是与广西株GX14YYA-J3的相似度高达96.5%~97.9%。通过种鸡精液ALV-J检测与gp85基因分析,为该地区鸡种的疾病防控提供了科学依据。 To determine the prevalence and molecular characteristics of J-subgroup avian leukosis virus(ALV-J)in local chicken breed of Longyan,a comprehensive analysis was conducted on 128 semen samples from these breeds.The samples were evaluated using an ELISA for the detection of p27 antigen and PCR for the identification of the ALV-J gp85 gene.The findings revealed that 17.2%of the samples exhibited a positive reaction for the p27 antigen.Subsequent PCR analysis confirmed the presence of ALV-J in 5.5%of the samples.Further genomic sequence analysis was performed to elucidate the genetic relatedness of the detected ALV-J strains to the reference strains of the J subgroup.The results demonstrated a high degree of nucleotide sequence identity,ranging from 92.0%to 97.9%.Notably,a particularly strong genetic affinity was observed between the Longyan strains and the Guangxi strain GX14YYA-J3,with sequence similarities reaching 96.5%to 97.9%.These findings underscore the genetic linkage between the circulating ALV-J strains in the Longyan region and the Guangxi strain,highlighting the significance of employing a combined ELISA and PCR diagnostic approach to enhance the accuracy of ALV-J detection in semen samples.Detection via the ALV-J antigen assay and analysis of the gp85 gene offers a scientific foundation for disease prevention and control among the chicken breed in the region.
