丹参抑制大鼠肝纤维化线粒体脂质过氧化 被引量:31

Inhibitory effect of Danshen on lipid peroxidation in mitochondria of hepatic fibrosis in rats

摘要 目的:研究活血化瘀中药的主药丹参不同浓度提取物对离体线粒体脂质过氧化的抑制作用.方法:提取肝纤维化模型鼠肝组织线粒体,配制不同浓度梯度丹参提取物反应液,以丙二醛(MDA)和过氧化物岐化酶(SOD)为指标,测定丹参对线粒体脂质过氧化抑制作用的量-效、时-效关系.结果:随丹参浓度增加,同一时刻内线粒体反应液MDA产生逐渐减少(反应20min时F=53.867,P=0.000;S-N-KP<0.05);SOD活性则逐渐升高(F=3.463,P=0.051;S-N-KP<0.05).随反应时间延长,相同浓度丹参的线粒体反应液中MDA增高同时SOD也逐渐增高(F=20.207,P=0.000;S-N-KP<0.05).反应的最强点发生在丹参浓度为2g/L或5g/L以及反应时间在15min或20min以后.结论:丹参对肝纤维化鼠肝脏线粒体脂质过氧化反应存在着一定的量-效与时-效关系. AIM:To study the anti-lipid peroxidation effect of Danshenroot extraction on mitochondria of hepatic fibrosis in rats.METHODS:The soluble ingredient of Danshen root was ex-tracted and prepared at different concentrations of reactionsolution. Taking malondialdehyde (MDA) and superoxidedismutase (SOD) as the test parameters, the authors deter-mined the dose-effect and time-effect relationships ofDanshen root extraction in anti-lipid peroxidation ofmitochondria.RESULTS:With the increase of concentrations of Danshenextraction, the MDA producing in mitochondria reaction so-lution wais decreased responsively at the same time, and,on the contrary, SOD was increased. The top point of reac-tion was reached at 2 mg/ml or 5 mg/ml of Danshen con-centrations and after 15 or 20 minutes from the beginningof reaction.CONCLUSION:There are dose- and time-effective relation-ships of Danshen root extraction on anti-lipid peroxidationof mitochondria of hepatic fibrosis in rats.
