甘草次酸对胃癌细胞SGC7901增殖的影响及其机制 被引量:12

Effect of glycyrrhetinic acid on proliferation of gastric cancer cell line SGC7901 and its mechanism

摘要 目的研究甘草次酸对胃癌细胞SGC7901增殖的影响及机制。方法人胃癌细胞SGC7901分为空白组、对照组和实验组,每组设置8个复孔,实验组细胞以100μmol·L-1甘草次酸处理,对照组以10μg·L-1阿霉素(DOX)处理,空白组细胞则以等量生理盐水处理。干预48 h后,于光学倒置显微镜下观察人胃癌细胞SGC7901形态。以噻唑蓝(MTT)及细胞克隆形成实验检测细胞增殖情况,以蛋白质印迹(WB)法检测细胞核因子-κB(NF-κB)信号通路蛋白表达情况。结果干预48 h后,空白组、对照组和实验组SGC7901细胞活率分别为(99.76±0.12)%,(56.64±7.37)%,(52.16±6.22)%,细胞克隆形成率分别为(54.65±3.32)%,(23.21±4.32)%,(22.43±3.56)%,细胞凋亡率分别为(2.43±0.21)%,(66.78±5.43)%,(69.62±4.37)%,p65蛋白相对表达量分别为1.06±0.21,0.63±0.15,0.66±0.24,p-IκB-α蛋白相对表达量分别为1.56±0.16,0.58±0.14,0.55±0.22,p-IKK-β蛋白相对表达量分别为0.96±0.14,0.45±0.08,0.48±0.15,对照组和实验组以上各指标与空白组比较,差异均有统计学意义(均P<0.05)。结论甘草次酸通过下调NF-κB信号通路蛋白表达抑制胃癌细胞SGC7901增殖,诱导其凋亡。 Objective To investigate the effect and mechanism of glycyrrhetinic acid on the proliferation of gastric cancer cell line SGC7901.Methods Human gastric cancer cell line SGC7901 was divided into blank group,control group and test group.Each group was treated with 8compound holes.The cells in test group were treated with 100μmol·L-1glycyrrhetinic acid,control group was treated with 10μg·L-1doxorubicin(DOX),and the cells in blank group were treated with the same amount of normal saline.After 48 h of intervention,the morphology of human gastric cancer cell line SGC7901 was observed under optical inverted microscope.MTT and clone formation assay were used to detect the cell proliferation.The expression of nuclear factor-κB(NF-κB)signaling pathway proteins were detected by Western blot(WB).Results After 48 h of intervention,the cell viability of SGC7901 cells in blank group,control group and test group were(99.76±0.12)%,(56.64±7.37)%,(52.16±6.22)%,the cell clone formation rates were(54.65±3.32)%,(23.21±4.32)%,(22.43±3.56)%,theapoptosis rates were(2.43±0.21)%,(66.78±5.43)%,(69.62±4.37)%,and the relative expression of p65protein were 1.06±0.21,0.63±0.15,0.66±0.24,the relative expression of p-IκB-αprotein were 1.56±0.16,0.58±0.14,0.55±0.22,the relative expression of p-IKK-βwere 0.96±0.14,0.45±0.08,0.48±0.15,compared with blank group,all indexes above in control group and test group were with significant difference(all P<0.05).Conclusion Glycyrrhetinic acid can inhibit the proliferation and induce apoptosis of gastric cancer cell SGC7901 by down-regulating the expression of NF-κB signaling pathway.
