拮抗菌Bs43的鉴定、抑菌机理及其对采后柑橘绿霉病的生防效果 被引量:13

摘要 【目的】为了开发对采后柑橘绿霉菌有效的生防措施,对从田间分离的1株生防细菌Bs43进行了鉴定,并对其拮抗活性、抑菌机理和生防效果进行了研究。【方法】运用BIOLOG鉴定板法及16S rDNA序列同源比对法对菌株Bs43进行鉴定;通过与其他柑橘采后病原真菌的对峙培养了解Bs43拮抗谱;通过含不同浓度代谢产物培养基培养柑橘绿霉菌,以及水-琼脂法、配合镜检研究Bs43代谢产物的拮抗活性及抑菌机理;用果实刺伤接种法对Bs43对采后柑橘绿霉病生防效果进行评价。【结果】菌株Bs43的生理生化特征及16S rDNA序列比对结果与解淀粉芽孢杆菌(Bacillus amyloliquefaciens)相符。该生防菌主要通过抑制孢子萌发、菌丝生长和产孢来发挥对柑橘绿霉菌的抑制作用。该生防菌有着较宽的拮抗谱,对其他柑橘采后病原真菌也具有良好的抑制活性,对柑橘绿霉菌的生防效果明显,且其抗菌物质的一部分对高温稳定。【结论】根据16S rDNA序列及生理生化特征,将菌株Bs43鉴定为解淀粉芽孢杆菌,综合菌株Bs43的拮抗谱、抑菌机理及生防效果,显示其具有一定的生防应用价值。 【Objective】Green mold caused by Penicillium digitatum is one of the main diseases in postharvest citrus during transportation and storage. To develop an effective biocontrol measure against postharvest citrus green mold,a bacterial biocontrol strain Bs43 isolated from rhizospheric soil in Jiangsu Province was identified and characterized,and its antagonistic activity,inhibitory mechanism and biocontrol effect were studied.【Method】Strain Bs43 was identified using both BIOLOG Gen Ⅲ plate and 16 S rDNA sequence homolog comparison. Its antagonistic spectrum was studied by culturing against 5 phytopatho-genic fungi of postharvest citrus diseases. Then the hyphal growth of P. digitatum cultured on elevated concentrations of fermentation filtrates and the spore germination cultured in 1% agar with elevated fermentation filtrates were observed to study antagonistic activity and inhibitory mechanism of strain Bs43.Finally,biocontrol effect against postharvest citrus green mold was assessed through stab inoculation.【Result】Strain Bs43 was identified as Bacillus amylolique faciens for its biochemical characteristics identified by BIOLOG Gen Ⅲ plate and 16 S rDNA sequence analysis. Its sequence was submitted to GenBank(accession number:KF914417). Strain Bs43 showed strong inhibition effect on P. digitatum spore germi-nation as well as a wide antagonistic spectrum against various phytopathogenic fungi,and a part of its inhibitory metabolites remained stable under heat stress.【Conclusion】Based on 16 S rDNA sequence analysis and its biochemical characteristics,strain Bs43 was identified as B. amyloliquefaciens. Strain Bs43 was approved to be an effective bio control agent by integrating assessment on its antagonistic spectrum,inhibitory mechanism and biocontrol effects against post harvest citrus green mold.
