摘要
【目的】明确甲氧香螨酯Scopoletin亚致死浓度对截形叶螨解毒酶活性的影响.【方法】采用叶片浸渍法测定了甲氧香螨酯对截形叶螨的亚致死浓度LC_(10)和LC_(30),并分析了用此两种浓度分别处理截形叶螨后2~72 h内共9个不同时间点的谷胱甘肽-S-转移酶(GSTs)、羧酸酯酶(CarEs)和多功能氧化酶(MFOs)的活性变化.【结果】LC_(30)浓度的甲氧香螨酯处理截形叶螨后,GSTs酶活性在2~72 h内显著高于对照,除48 h外显著高于LC_(10)处理;LC_(10)浓度的甲氧香螨酯处理截形叶螨后,GSTs酶活性在24~60 h内显著高于对照,其他时间点与对照差异不明显;LC_(30)浓度的甲氧香螨酯处理截形叶螨后,CarEs酶活性2~72 h内显著高于对照,在24~48 h内显著高于LC_(10)处理,其他时间点差异不显著;LC_(10)浓度的甲氧香螨酯处理截形叶螨后,CarEs酶活性在2~72 h内显著高于对照;LC_(30)浓度的甲氧香螨酯处理截形叶螨后,MFOs酶活性在2、36~48和72 h显著高于对照,在6、24 h显著低于对照,4、12、60 h与对照无明显差异,在2、6、24 h显著低于LC_(10)处理,其他时间点与LC_(10)处理无明显差异;LC_(10)浓度的甲氧香螨酯处理截形叶螨后,MFOs酶活性在2、24、48、72 h显著高于对照,6 h显著低于对照,其他时间点与对照无明显差异.【结论】GSTs、CarEs可能参与了甲氧香螨酯的代谢,MFOs的作用还需进一步研究.
【Objective】 To clarify the effect of sublethal concentration of scopoletin on activities of detoxification enzymes of Tetranychus truncates.【Method】 The sublethal concentration of scopoletin(LC10 and LC30) on the female adult mite of T.truncates were tested by the immerse leafs method,and the activity of glutathione-S-transferase(GSTs),carboxylesterase(CarEs) and MFOs in T.truncates were analyzed by ELISA Kit during 2~72 h.【Result】The activity of GSTs treated with LC30 scopoletin was significantly higher than the control in 2~72 h and higher than LC10 except 48 h,the activity of GSTs treated with LC10 scopoletin was significantly higher than the control in 24~60 h except others.The activity of CarEs treated with LC30 scopoletin was significantly higher than the control in 2~72 h and higher than LC10 in 24~48 h,the activity of CarEs treated with LC10 scopoletin was significantly higher than the control in 2-72 h.The activity of MFOs treated with LC30 scopoletin was significantly higher than the control in 2 h,36~48 h and 72 h,significantly lower than the control in 6 h and 24 h and others no significant difference and significantly lower than the LC10 in 2 h,6 h and 24 h and others no significant difference,the activity of MFOs treated with LC10 scopoletin was significantly higher than higher than the control in 2 h,24 h,48 h and 72 h,significantly lower than the control in 6 h and others no significant difference.【Conclusion】 GSTs and CarEs may be involved in the metabolism of scopoletin in T.truncates,and the role of MFOs needs further study.
