摘要
目的检测脑红蛋白(Ngb)在视网膜母细胞瘤(RB)中的表达,探讨Ngb在RB发生、发展机制中的意义。方法使用免疫组化方法 SP法检测Ngb在RB瘤体中央、瘤体边缘和肿瘤未侵及视网膜中的表达,并与神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)的表达进行对比分析;比较Ngb在分化型和未分化型RB中的表达差异。结果 Ngb在肿瘤未侵及视网膜的色素上皮层、感光细胞内节、内颗粒层、节细胞层及内外丛状层均有表达;Ngb在RB瘤体的表达总阳性率为81.3%,与NSE,GFAP的表达比较差异有统计意义(瘤体中央,χ2=25.828,P<0.05;瘤体边缘,χ2=12.150,P<0.05);Ngb在"菊形团"细胞和血管周围肿瘤细胞的表达较为广泛,但在分化型与未分化型中的表达无明显差异。结论 RB可以生成一定的Ngb,说明肿瘤细胞具有视网膜神经元的某些特征,但Ngb在RB的发生、发展中作用较局限。
Objective To study the expression of Ngb in RB,and investigate Ngb's roles in the development of RB. Methods Expression of Ngb was evaluated by immunhistochemistry in the central region,the marginal region and the normal retinal tissue of RB as well as analysed the correlation among Ngb,NSE and GFAP; and compared the differences of Ngb in differentiated and undifferentiated type of RB. Results Ngb expressed in the normal retinal tissue as well as in the pigment epithelium layer,photoreceptor inner segments,inner and outer plexiform layers,inner nuclear layer,and ganglion cell layer. The total positive rate of Ngb in RB was 81.3%. In the central region and the marginal region of RB,the expression levels of Ngb,NSE and GFAP were significantly different( χ2= 25.828,P<0.05 and χ2= 12.150,P<0.05). Cells of rosettes and surrounding blood vessels showed a tendency to be stained with Ngb,but there was no obvious difference between differentiated and undifferentiated type. Conclusion RB can generate a certain number of Ngb. It proves that tumor cells have some characteristics of retinal neurons,but the function of Ngb in the development of RB is limited.
