摘要
本文应用细胞因子依赖细胞株CTLL和7TD1及。<sup>3</sup>H-TDR掺入方法,分别对6种常用培养杂交瘤的饲养细胞培养上清和条件培养液中IL-2和IL-6的生物学活性水平进行了定量检测和比较。结果发现,除大鼠混合胸腺细胞培养上清中含有约1u/ml IL-2外,其它均未测出IL-2的生物学活性。但6种上清中都含有IL-6,其中人成纤维细胞CRL<sub>1506</sub>和人内皮细胞中含有很高水平的IL-6,分别为10000u/ml和2512u/ml;大鼠混合胸腺细胞和小鼠腹腔巨噬细胞培养上清分别为158u/ml和126u/ml;小鼠脾脏细胞和胸腺细胞培养上清中IL-6相对较低,分别为79u/ml和16u/ml。此外,本文还对常用饲养细胞及条件培养液在杂交瘤细胞生长中可能起的主要作用进行了讨论。
The IL-2 and IL-6 biological activities in feeder cell culture supernatants,or conditioned media for hybridoma technique were detected by cytokine dependent cell 3H-TdR incorporation methods. The results indicated that these samples contained no detectable IL-2 except supernatants from Rat mixed thymocyte culture and the levels of IL-6 activity were 10 000u/ml, 2 512u/ml, 158u/ml, 126u/ml, 79u/ml and 16u/ml in supernatants from CRL1506, endotheliocyte, Rat mixed thymocyte, macrophage, splen-ocyte and thymocyte culture, respectively. The main role of feeder cells and conditioned media for hybridoma technique was discussed.
