摘要
DNA在生命活动中具有重要意义,针对DNA检测技术日益增多,近些年一种无酶等温高效的超灵敏扩增技术,杂交链反应(HCR)逐渐引起广泛的关注。HCR,仅在特定的核苷酸引发物(NI)和两个特殊设计的发夹DNA(H1,H2)同时存在的情况下进行自组装,实现了原位扩增反应。由于NI,H1,H2的可编辑性及可修饰性,使得HCR适用于分子生物学的各个领域,具有广泛的应用前景。该文通过HCR在核酸检测、蛋白检测、细胞检测及生物成像等方面的应用进展来探究其优点和不足。
DNA has great significance in life activities,and the technology of DNA detection is increasing.Recently,hybridization chain reaction(HCR),an enzyme-free isothermal ultra-sensitive amplification technology,has gradually attracted widespread attention.HCR,self-assembly only in the presence of a specific nucleotide initiator(NI)and two specially designed hairpin DNA(H1,H2),achieves an in situ amplification reaction.With the editability and modification of NI,H1 and H2,HCR can be used to various fields of molecular biology,and has broad application prospects.In this paper,we explore the application of nucleic acid detection,protein detection,cell detection and bioimaging and compare the advantages and disadvantages in HCR.
