摘要
叶绿素是调节光合作用的关键色素,对籽粒形成有着重要作用。本研究以美国半矮秆大豆Charleston为母本,东北地区主栽品种东农594为父本杂交衍生的147个重组自交系群体为材料,基于经SLAF测序获得的大豆高密度遗传图谱,利用复合区间作图法(CIM)、多重区间作图法(MIM)和完备区间作图法(ICIM)对大豆叶绿素含量进行QTL联合定位分析,并结合大豆基因组基因注释信息对QTL区段内的候选基因进行预测。利用CIM算法定位出2个QTL,表型遗传贡献率分别为6%和9.3%。利用MIM算法定位到了1个QTL,表型遗传贡献率为8.1%。利用ICIM算法定位到了1个QTL,表型遗传贡献率为7.76%。其中qchl-G-1被CIM和MIM两种算法同时检测到。在上述3个QTL区段内共含有151个基因,根据大豆基因组基因注释信息,筛选到了3个与叶绿素相关的候选基因,这些结果为叶绿素含量的遗传剖析和标记辅助育种提供理论基础,有利于分子辅助育种的发展。
Chlorophyll is the key pigment of regulating photosynthesis and plays an important role in grain formation. In this study, we used 147 recombination inbred lines(RILs) derived from a cross of the semi-dwarf soybeans Charleston(♀) from the United States and Dongnong 594(♂) which was the main variety in northeast Chinese experimental materials. We based on the soybean high-density genetic map by SLAF sequencing technology, used CIM, MIM and ICIM methods to QTL joint positioning analyze the chlorophyll content of soybean and predicted the candidate gene in the QTL section by combining with the soybean genome gene annotation information. We used CIM method to map two QTLs whose phenotypic genetic contribution rates were 6% and 9.3%. We used MIM method to map one QTL whose phenotypic genetic contribution rate was 8.1%. We used ICIM method to map one QTL whose phenotypic genetic contribution rate was 7.76%. qchl-G- 1 was identified by CIM and MIM method. There were 151 genes among the above three QTL segments. We screened out 3 candidate genes associated with chlorophyll ba sed on the soybean genome annotation information. These results would provide theoretical basis for the genetic analysis of chlorophyll and marker assisted breeding, and would be beneficial to the development of marker assisted breeding.
