摘要
利用对流分配、硅胶柱层析、薄层层析、中压液相层析等分离技术 ,从基因工程菌 WSJ- 1的活性产物生技霉素中分离了 16个化合物。其中除主要产物生技霉素 A1 、B1 、E1 (4″- O-异戊酰螺旋霉素 、 、 )外 ,还包括生技霉素 A2α,A2β,生技霉素 B3,C1 (4″- O-丁酰 ,异丁酰 ,丙酰 ,乙酰螺旋霉素 )及生技霉素 B2α,B2β,C2 ,D1 (4″- O-丁酰 ,异丁酰 ,丙酰 ,乙酰螺旋霉素 ) ,以及新的 4″-酰化螺旋霉素生技霉素 B0 [4″- O- (3-甲基 - 2 -丁烯酰基 ) -螺旋霉素 ]。还有可能是 4″- O-酰化螺旋霉素生物合成中间体的生技霉素 A0 (普拉特霉素 A1 ) ,生技霉素 P1 、P2 (螺旋霉素 , )和生技霉素 P3(新螺旋霉素 )。文中报道了这些化合物的分离 ,性质及结构。并总结报道了具有重要鉴别意义的这些化合物中 C- 3,C- 4″酰基侧链的 1 H及 1 3C- NMR数据。
16 components were isolated from shengjimycin complex, a group of hybrid antibiotics produced by a genetically engineered strain WSJ 1. Besides the major components: shengjimycin A 1, B 1 and E(4″ O isovaleryl spiramycin Ⅲ,Ⅱ and Ⅰ), shengjimycin A 2α , A 2β , shengjimycin B 3,C 1(4″ O butyryl, isobutyryl, propionyl, acetyl spiramycin Ⅲ), shengjimycin B 2α , B 2β , C 2, D 1(4″ O butyryl, isobutyryl, propionyl, acetyl spiramycin Ⅱ), new compound 4″ O (3 methyl butenoyl) spiramycin Ⅱ, compounds platenomycin A 1 (possibly intermediates of 4″ O acyl spiramycins biosynthesis), spiramycin Ⅲ, Ⅱ and neo spiramycin Ⅲ were also isolated and structurally identified. In this paper, isolation, characteristics, and structures of these components were reported. 1H and 13 C NMR data of C 3 and C 4″ acyl side chain of these compounds had also been summarized and reported.
