摘要
以菊苣叶片、茎段、花蕾和花瓣为外植体 ,进行离体培养试验 ,结果表明 ,叶片、茎段、花蕾可产生淡黄色的愈伤组织 ,并可诱导出不定芽 ,出愈率为 10 0 % ,叶片和茎段的分化率为 10 0 %。诱导愈伤组织及分化的最佳培养基为 :MS+BA2 mg/ L+IBA0 .3mg/ L;生根培养基为 :1/ 2 MS+NAA0 .1mg/ L,生根率为 10 0 %。
The tissue culture and plant regeneration of Cichorium intybus L. cv. Puna was carried out using leaf, stem, flower bud and petal as explants, respectively. The result indicated that some pale yellow callus could be induced and adventitious buds could be differentiated from explants of leaf, stem and flower bud, respectively. The optimum medium for callus induction and bud differentiation was MS+BA 2 mg+IBA 0.3 mg/L; The optimum medium for root formation was 1/2 MS+NAA 0.1 mg/L; the rate of callus induction, bud differentiation and root formation was 100%, respectively.
