摘要
使用根据转基因Bt176玉米中的外源基因CryIA(b)和内源基因Zein设计的引物和TaqMan荧光探针和ABIPRISM 770 0定量PCR仪对玉米粉中Bt176玉米的含量进行了定量检测 ,建立了Bt176玉米参照样品CryIA(b)和Zein的Ct值之差与样品中Bt176玉米含量之间的标准曲线和线性回归方程 。
The genetically modified Bt176 maize was quantitatively detected by ABI PRISM 7700 sequence detector with the PCR primers and TaqMan fluorescence probes designed according to the sequences of endogenous Zein gene and exogenous CryIA(b) gene (Table 1). The Zein gene and CryIA(b) gene was amplied respectively (Figs.1 and 2), The standard curve of ΔCt between Zein gene and CryIA(b) gene vs the Bt176 content in reference materials was generated and a linear regression equation was obtained (Table 2, Fig.3) by which the Bt176 maize content in two maize powder samples was detected (Table 3).
