中国藏族与维吾尔族Y染色体DYF155S1基因座的遗传多态性与民族差别 被引量:1

摘要 目的 :探讨Y染色体DYF15 5S1基因座的遗传多态性及民族间差异。方法 :应用小卫星可变重复序列PCR荧光显谱及DNA序列测定技术 ,对 12 2例中国藏族和维吾尔族男性个体的DYF15 5S1基因座的多态性进行检测。结果 :共检出 5种重复序列类型 ,包括 1型、3型、4型与新的 7型和未知型 ,7型是在 3型的基础上T2 2A置换所形成 ,在 72例藏族群体中发现 18例 ,维吾尔族群体中未检出 ,可作为民族特征性遗传标记。重复序列的排列方式以 3 13 4顺序多见 ,在藏族和维吾尔族群体中各占 61.1%和 48.0 % ,是黄种人的特点。 13 4顺序在维吾尔族群体中占第二位 ,为 3 2 .0 % ,3′端的 4型重复序列的平均数目在维吾尔族群体为 18.0条 ,明显高于藏族群体的 13 .3条 ,显示出维吾尔族群体有接近白种人的特点 ,同时存在黄种人与白种人混杂的痕迹。藏族群体的 2 5 .0 %为73 13 4排列。结论 :DYF15 5S1基因座具有非常高的遗传多态性和明显的民族差异 。 Objective: We tried to understand the genetic polymorphism and population difference of locus DYF155S1 on human Y Chromosome. Methods: Using minisatellite variant repeat PCR(MVR PCR), automated fluorescence detection, DNA sequence analysis, we studied the genetic polymorphism of locus DYF155S1 in total 122 individuals from Zang and Uygur nationality. Results: We detected 5 types of repeat units, that is type 1,type 3,type 4,type 7 and type unknown. Type 7 of which 18 out of 72 samples were observed in the Zang nationality was resulted from the T22A substitution on type 3, and was not detected in the Uygur nationality; therefore type 7 may be regarded as a racial characteristic genetic marker. The common arrangement of the repeat units was 3134,that was detected at 61.1 % and 48.0% respectively in the Zang nationality and the Uygur nationality. The abundance of arrangement 3134 was a character of yellow race. Next to arrangement 3134 was arrangement 134 which was detected at 32.0% in the Uygur nationality. The average number of type 4 repeat unit to the 3′ terminal was 18.0 in the Uygur nationality, which was much more than13.3in theZang nationality. That implied the Uygur nationality might own the character close to the white race as well as the mixed trail of yellow race and white race. The arrangement 73134 was detected at 25.0% in the Zang nationality. Conclusion: DYF155S1 locus has great genetic polymorphism and obvious population difference, and its significance should be paid more attention in the forensic science and human genetics research.
