摘要
目的 为探讨炎性细胞因子 (CK)在Alzheimer病 (AD)慢性炎性病理过程中的作用。方法 本研究采用ELISA法对AD病人 (1 1例 )、血管性痴呆 (VD)病人 (1 3例 )与正常对照 (1 3名 )的血清与脑脊液 (CSF)、白细胞介素 1 β(IL 1 β)和肿瘤坏死因子 (TNF α)的水平进行了测定。 结果 与VD(38 51± 1 4 39)pg/ml、正常对照 (33 98± 1 6 31 )pg/ml相比 ,AD病人血清IL 1 β水平 (60 1 3± 1 7 81 )pg/ml明显升高 ,P <0 0 1。AD病人血清TNF α水平 (81 2 0± 2 3 99)pg/ml虽较VD(71 77± 2 8 86)pg/ml和正常对照 (73 91± 1 7 69)pg/ml轻度升高 ,但并无统计学意义。AD病人CSFIL 1 β水平 (38 2 5±1 8 42 )pg/ml明显高于对照组 (2 0 2 5± 1 5 86 )pg/ml和VD病人 (32 1 8± 1 9 81 )pg/ml,P <0 0 5。AD病人CSFTNF α水平 (1 4 2 58± 1 1 46)pg/ml也明显高于VD病人 (1 1 5 46± 1 9 52 )pg/ml,P <0 0 5 )和对照组 (1 0 6 2 3± 2 6 1 3)pg/ml,P <0 0 1。结论 上述结果表明 ,炎性细胞因子IL 1 β和TNF α在AD的神经变性过程起着重要的作用 ,炎症可参与AD的发病机制。
Objective To investigate the levels of interleukin 1β (IL 1β) and tumor necrosis factor α (TNF α) in both serum and CSF in patients with Alzheimer's disease Methods Eleven patients with Alzheimer's disease,13 patients with vascular dementia (VD) and 13 healthy controls were analyzed by using commercial ELISA kits Results The average serum level of IL 1β was found higher in AD patients(60 13±17 81 pg/ml) than in VD patients(38 51±14 39 pg/ml) and in healthy controls(33 98±16 31 pg/ml)( P <0 01, respectively) Though the serum TNF α level was slightly higher in AD patients(81 20±23 99 pg/ml)than in VD patients(71 77±28 86 pg/ml)or healthy controls(73 91±17 69 pg/ml), none reached at a statistical significance CSF IL 1β levels were found elevated in AD patients(38 25±18 42 pg/ml)as compared to the healthy controls(20 25±15 86 pg/ml)and VD patients(32 18±19 81pg/ml)( P <0 05, respectively) AD patients were found to have higher CSF TNF α levels(142 58±11 46 pg/ml)than VD patients(115 46±19 52 pg/ml)and controls(106 23±26 13 pg/ml)( P <0 05 and P <0 01, respectively) Conclusion These results strongly suggest that inflammatory cytokines IL 1β and TNF α should be important factors in the neurodegeneration process associated with AD and the inflammation should significantly contribute to the pathogenesis of AD Specific therapeutic strategies aimed at modulation of cytokines network in AD would have considerable promise
