摘要
为了研究甘露醇在猪精液冷冻过程中对精子品质的影响,试验分别在猪精液中添加0,1,2,3 mg/mL甘露醇,采用精子动(静)态图像检测系统(CASA)检测精子活力,采用低渗肿胀法检测精子质膜的完整性,采用考马斯亮蓝染色法检测精子顶体膜的完整性,对精子凋亡基因的表达量进行检测,并通过体外受精后精子的黏附程度进一步验证。结果表明:在猪精液中分别添加1,2 mg/mL甘露醇,精液冷冻-解冻后精子的质量均显著改善(P<0.05),其中添加2 mg/mL甘露醇时精子内凋亡基因表达量最少。说明甘露醇在冷冻保存过程中对精子具有保护作用,且最适添加浓度为2 mg/mL。
This experiment was performed in four groups,the semen in the test groups were added 0,1,2 and 3 mg/mL mannitol,respectively.Computer-Aided Sperm Analysis(CASA)was used to detect sperm motility,the integrity of sperm plasma membrane was detected by Hypotonic swelling test(HOST),the integrity of sperm acrosome membrane was detected by Coomassie brilliant blue staining,and the expression of sperm apoptotic gene were detected.Finally,sperm-oocyte binding ability was tested by in vitro fertilization method.The results are as follows:After freezing and thawing of the treatment group with 1,2 mg/mL mannitol in the cryopreservation agent,the sperm quality was significantly improved compared with the control group(P<0.05),the 2 mg/mL mannitol treated group have the lowest sperm expression of apoptotic genes in sperm.It indicates that mannitol has a protective effect on sperm cryopreservation,and the best concentration is 2 mg/mL during cryopreservation.
