氢化荧光素二乙酸酯的合成及其在活细胞染色中的应用

摘要 目的:合成氢化荧光素二乙酸酯,并用于肿瘤细胞的染色标记。方法:通过锌粉和冰醋酸还原荧光素制备二氢荧光素,再经乙酸酐酰化合成氢化荧光素二乙酸酯。氢化荧光素二乙酸酯对肿瘤细胞进行染色,并与荧光素二乙酸酯的染色结果进行对照。结果:不论是贴壁还是悬浮细胞,当浓度为1.0 mg/L孵育染色5 min时,FHDA可基本标记细胞,背景低,可定量检测细胞的荧光强度。结论:氢化荧光素二乙酸酯可用于肿瘤细胞的荧光标记及活性氧等细胞活性物质的检测。 Objective Synthesis of fluorescindiacetate and staining cancer cells .Methods Fluorescein was reduced to fluorescin by zinc powder and ice acetic acid , fluorescindiacetate was prepared by acetation of fluoresce-in with acetic anhydride .Cancer cells were stained with fluorescindiacetate and campared with the results that stained with fluoresceindiacetate .Results Adherent cells and suspension cells could be all stain when incubating and staining 5 mins with 1.0 mg/Lfluorescindiacetate.Fluorescent intensity in cells could be observed because of low fluorescnet level in the background .Conclusion Fluorescindiacetate could be used for staining cancer cell and detecting cell active substances such as active oxygens .
