摘要
目的 :探讨低氧诱导因子-1α(HIF-1α)、基质金属蛋白酶抑制剂-1(TIMP-1)在非小细胞肺癌(NSCLC)中的表达及意义。方法 :采用免疫组化法检测96例NSCLC及癌旁正常组织中HIF-1α、TIMP-1的表达,并分析二者与临床病理参数的关系。结果 :HIF-1α在NSCLC(80/96)中的阳性表达显著高于癌旁正常组织(28/96),TIMP-1在NSCLC(71/96)中的阳性表达显著高于癌旁正常组织(15/96)。低分化中HIF-1α、TIMP-1阳性表达率显著高于癌旁正常组织,Ⅲ-Ⅳ期中HIF-1α、TIMP-1阳性表达率显著高于Ⅰ-Ⅱ期,有淋巴结转移HIF-1α、TIMP-1阳性表达率显著高于无淋巴结转移。而NSCLC中HIF-1α、TIMP-1阳性表达率与年龄、性别、肿瘤大小、吸烟史、病理类型无关。结论 :HIF-1α、TIMP-1在NSCLC的发生、侵袭、转移起重要作用,可能成为NSCLC临床治疗的靶点之一。
Objective To explore significance and expression of hypoxia inducible factor-1α(HIF-1α) and tissue inhibitor of metalloproteinase-1(TIMP-1) in non-small cell lung cancer(NSCLC). Method The expression of HIF-1α and TIMP-1 in 96 cases of NSCLC and adjacent normal tissues was detected by immunohistochemical method. The relationship between HIF-1α, TIMP-1 and clinical pathological parameters was analyzed. Result The positive rate of HIF-1αin NSCLC(80/96) was higher than that in adjacent normal tissues(28/96). The positive rate of TIMP-1 in NSCLC(71/96) was higher than that in adjacent normal tissues(15/96). The positive rate of HIF-1α and TIMP-1 in poorly differentiated was higher that in the high and moderately differentiated. The positive rate of HIF-1α and TIMP-1 in Ⅲ-Ⅳ stage was higher than that in Ⅰ-Ⅱ stage. The positive rate of HIF-1α and TIMP-1 in lymph node metastasis was higher than that in without lymph node metastasis. The expression of HIF-1α was positively correlated with the expression of TIMP-1 in NSCLC. Conclusion HIF-1α and TIMP-1 played an important role in the occurrence, invasion and metastasis of NSCLC, and might be one of the targets for the clinical treatment of NSCLC.
