摘要
目的:观察胶质瘤致病相关蛋白1(GLIPR1)对肺癌A549细胞生长及分化的影响,以期为肺癌药物的研制提供线索。方法:构建重组慢病毒Lenti-GLIPR1和包含GLIPR1胞外可溶性区段(GLIPR1-S)基因片段的LentiGLIPR1-S,感染A549细胞后,通过Western blot法检测GLIPR1或GLIPR1-S蛋白的表达,细胞计数法检测细胞生长,免疫细胞化学法观察细胞分化。结果:A549细胞经慢病毒Lenti-GLIPR1和Lenti-GLIPR1-S感染后,均可表达相应蛋白,细胞生长受抑,细胞形态发生改变,向肺上皮细胞分化。结论:感染GLIPR1和GLIPR1-S均可促进A549细胞向肺上皮细胞分化,抑制A549细胞的生长。
Aim:To explore the effects of glioma pathogenesis-related protein 1(GLIPR1) on cell growth and differentiation of A549 cells for providing a potential therapeutic agent for lung cancer therapy.Methods:Recombinant lentivirus Lenti-GLIPR1 and Lenti-GLIPR1-S were constructed and infected A549 cells.The expressions of GLIPR1 or GLIPR1-S were detected by Western blot,and cell counting was performed.Immunocytochemical staining was used to analyze the differentiation of A549 cells.Results:A549 cells could express GLIPR1 or GLIPR1-S proteins after being infected by LentiGLIPR1 or Lenti-GLIPR1-S.GLIPR1 and GLIPR1-S infection could both inhibit A549 cell growth,change the shape of A549 cells,and promote it differentiating to lung epithelial cells.Conclusion:Both GLIPR1 and GLIPR1-S infection could inhibit A549 cell growth,and promote them to differentiate to lung epithelial cells.
