摘要
目的:对不同产地松叶药材的质量进行评价,建立松叶药材的指纹图谱,完善松叶药材的质量评价系统,为其资源的开发利用提供科学依据。方法:收集15批不同产地的松叶药材,采用薄层色谱法进行定性鉴别;采用高效液相色谱法进行定量鉴别,并对水分、水溶性浸出物进行测定;同时建立松叶的高效液相色谱指纹图谱。结果:薄层色谱法斑点清晰,分离度好;采用HPLC法对松叶进行含量测定,经方法学考察验证,槲皮素与山奈素分别在35. 34~176. 7ng和71. 74~358. 7ng范围内呈良好的线性关系,平均加样率分别为96. 2%、97. 16%(RSD分别为1. 81%、1. 66%,n=6),说明该方法灵敏可行,重现性好;建立了松叶的指纹图谱,并对15批松叶样品的指纹图谱进行了相似度评价。结论:本研究建立的方法可行、简便,可为松叶质量标准的建立提供参考依据。
Objective:To evaluate the quality of pine needles from different producing areas,to establish the fingerprint for pine needles,to perfect the quality assessment system for pine needles,and to provide a scientific basis for the exploitation and utilization of pine needle resources.Methods:A total of 15 batches of pine needles from different producing areas were collected,and thin-layer chromatography(TLC)was used for qualitative identification.High-performance liquid chromatography(HPLC)was used for quantitative identification,and water content and water-soluble extract were measured.The HPLC fingerprint of pine needles was established.Results:Clear and well separated spots were observed on TLC.HPLC was used for content determination of pine needles;after methodological examination and validation,quercetin showed a good linear relationship within the range of 35.34~176.7 ng,with an average recovery rate of 96.2%(relative standard deviation[RSD]=1.81%,n=6),and kaempferide showed a good linear relationship within the range of71.74~358.7 ng,with an average recovery rate of 97.16%(RSD=1.66%,n=6).These results suggested that this method was sensitive and feasible and had good repeatability.The fingerprint was established for pine needles,and the similarity of fingerprint was evaluated for the 15 batches of pine needle samples.Conclusion:The method established in this study is simple,convenient,and feasible and can provide a reference for establishing quality standard for pine needles.
