摘要 目的美洲大蠊药效物质提取工艺优化。方法以得率及体外抗肿瘤细胞活性为评价指标,筛选美洲大蠊药效物质的提取溶剂;以得率及体外抗肿瘤细胞活性为评价指标,以溶剂剂量、提取时间、浸泡时间为影响因素,正交试验优化美洲大蠊药效物质的提取工艺。结果美洲大蠊药效物质最佳提取溶剂为75%乙醇,其IC_(50)为9.11 mg/mL;美洲大蠊药效物质最佳提取工艺为溶剂剂量15倍、提取时间1.0小时、浸泡时间2.0小时。结论筛选美洲大蠊药效物质的最佳提取工艺,为美洲大蠊提取物为主的制剂开发提供理论依据。 Objective To optimize the extraction technology for the effective substance of Periplaneta americana. Methods Taking the yield and in vitro antitumor activity as evaluation indexes, the extraction solvent for Periplaneta americana was selected. Taking yield and in vitro antitumor activity as evaluation indexes, orthogonal design was used to optimize the extraction process of effective substances from Periplaneta americana with solvent dosage, extraction time and soaking time were taken as influence factors.Results The best extraction solvent for Periplaneta americana was 75% ethanol of which the IC50 was 9.11 mg/mL. The optimum extraction process of effective substances from Periplaneta americana was 15 times of solvent dosage, 1.0 h of extraction time and 2.0 h of soaking time. Conclusion The optimum extraction process of the effective substances from Periplaneta americana is selected, which provides a theoretical basis for the development of Periplaneta americana extract.
