摘要
目的 :为观察tPNS对HCS刺激的RASMC增殖及其释放NO的影响。方法 :运用兔HCS刺激体外培养的RASMC而形成的细胞异常增殖为模型 ,通过MTT法、电镜观察和Green法 ,观察了tPNS对RASMC的保护作用及其对NO释放的影响。结果 :HCS可明显抑制NO的释放、刺激RASMC增殖并导致其表型的改变 ,而tPNS可诱导NO的释放 ,明显抑制HCS对RASMC的刺激增殖作用 ,与HCS组相比具有显著差异 (P <0 .0 1)。结论 :tPNS抗动脉粥样硬化作用可能部分是通过调节NO的合成而抑制RASMC增殖来实现的。
Objective:To investigated the protective effect on the proliferation of cultured RASMC and on the release of NO from RASMC affected by the tPNS for understanding the pharmacological mechanism of tPNS.Methods:The protective effect of tPNS on the proliferation of RASMC was observed by measuring MTT metabolism and observing cell ultrastructure under TEM, and on the NO amounts from RASMC were determined by the methods of Green. Results:HCS increased MTr metabolism of RASMC and caused change of phenotype and reduced the release of NO from RASMC. However, tPNS can inhibit MTT metabolism of RASMC stimulated by HCS and induce the release of NO from RASMC and raised the NO amounts. The results were significant compared with HCS group( P <0.01).Conclusion:The tPNS may prevent atherosclerosis and inhibit atherogenesis progression by inducing the release of NO from RASMC and interfering with the proliferation of RASMC.
