摘要
目的 :探讨大鼠海马神经元血红素氧合酶 1(hemeoxygenase 1,HO 1)在正常发育期的表达及在高热惊厥时的变化。方法 :发育期大鼠随机分为 2组 :正常组 (n =6 )和高热处理组 ,根据大鼠是否惊厥 ,后者又分为高热未惊厥组 (n =6 )和高热惊厥组 (n =6 )。利用热水浴模型诱导大鼠高热惊厥 ,选择不同鼠龄的发育期大鼠 ,利用免疫组织化学方法 ,观察大鼠海马神经元HO 1表达的变化。结果 :(1)出生 3h的新生大鼠海马HO 1未见表达 ,生后 1周表达水平较高 ,生后 2周达高峰 ,3周表达降低 ,生后 6周表达明显降低 ,成熟后 (2月 )表达维持在较低水平。(2 )海马CA1、CA3和门区神经元HO 1表达光密度的比较 :高热惊厥组HO 1表达增加 ,与正常组及高热未惊厥组比较 ,差异均有显著性 (P <0 .0 1) ,高热未惊厥组与正常组比较差异无显著性 (P >0 .0 5 )。结论 :不同鼠龄的发育期大鼠 ,海马神经元HO 1表达水平不同 ;发育期大鼠高热惊厥诱导海马神经元HO 1表达显著增加 ,这种表达增加可能与高热惊厥脑损伤有关。
Objective: To explore the change of heme oxygenase 1( HO 1) expression of the hippocampal neurons in immature rats during their normal development, and the influence of febrile convulsion on HO 1 expression. Methods: Developing rats were divided randomly into normal control group( n =6) and hyperthermia treated group, which was further divided into hyperthermic group(non convulsive, n =6) and febrile convulsion(FC) group( n =6), according to whether they experienced febrile seizure. The seizures were induced by warm water bath. An immunohistochemical method was adopted to explore the HO 1 expression in hippocampal neurons. Results: (1)The normal development of HO 1 expression: no expression of HO 1 was observed in the newborn(3 hours after birth)rats. Positive labeling was relatively high 1 week after birth, and became most obvious 2 weeks after birth, and gradually decreased thereafter, leaving a low level in mature rats(2 months after birth). (2)The immunohistochemical expression of HO 1 in FC, hyperthermic and normal control rats: light density of HO 1 expression in hippcampal CA1,CA3 and hile neurons of rats was detected. A significant difference( P <0.01) was demonstrated among normal control, hyperthemic and FC groups. Light density in FC group was significantly different from that in normal and hyperthemic groups( P <0.01).No significant difference was observed in the hyperthemic group compared withthat in normal group( P >0.05). Conclusion: HO 1 expression of hippocampal neurons in rats was at different levels during different growth processes. FC induced high expression of HO 1 in hippocampal neurons in developing rats. The results in the present study suggest that increased HO 1 expression may be one of the mechanisms of hippocampal neuron damage in developing rats.
