摘要
以烟草叶片为外植体,通过根癌农杆菌叶盘转化法,将番茄Pti5-VP16基因导入烟草中。经卡那霉素筛选,获得了27株卡那霉素抗性植株,经PCR扩增和Southern杂交分析,证明抗性植株中整合了Pti5-VP16基因,表明所构建的含Pti5-VP16基因的植物表达载体pBI121UCH1具有正确的阅读框,并能在异源植物中表达。
Leaf segments of tobacco were infected by Agrobacterium tumefaciens EHA105 with plasmid pBI121UCHl ,and then 27 kanamycin resistant plants were obtained. PCR and Southern analysis proved that Pti5-VPl6 gene has already integrated into the genome of the tobacco,and the expression vector has a right open read frame that can express in heterologous plants.
