摘要
目的 :研究钾通道阻断剂、一氧化氮 (NO)对继代培养大鼠肺动脉平滑肌细胞 (PASMC)钾电流 (IK)的作用。 方法 :使用全细胞膜片钳技术观察继代培养大鼠 PASMC IK 及钾通道阻断剂格列本脲 (Gli) ,四乙胺 (TEA) ,4 -氨基吡啶 (4- AP)和 NO供体 S-亚硝基乙酰青霉胺 (SNAP)对其的作用。 结果 :继代培养大鼠 PASMC较急性分离者之 IK 明显降低。 4 - AP和高浓度TEA显著降低继代培养大鼠 PASMC之 IK,SNAP显著增加继代培养大鼠 PASMC之 IK。结论 :继代培养大鼠 PASMC钾通道特性发生改变 ,但通道阻断剂和 NO仍能显著改变其活性。
Objective: To study the effect of potassium channel inhibitor and nitric oxide(NO) on the potassium current of sub cultured rat pulmonary artery smooth muscle cells(PASMC). Methods: The potassium current (I K) of sub cultured rat PASMC and the effect of potassium channel inhibitor Gli, TEA, 4 AP, and NO donor SNAP on it were observed by whole cell patch clamp techniques. Results: The I K of sub cultured rat PASMC was remarkably reduced than that of the acute dispersed ones. 4 AP and high concentration TEA remarkably reduced I K of sub cultured rat PASMC, while SNAP remarkably increased the I K. Conclusion: The character of sub cultured rat PASMC potassium channel is remarkably changed, but the potassium channel inhibitors and NO can still change its activity remarkably.
