摘要
目的研究逆转录酶抑制剂(AZT)对卵巢癌细胞系HO-8910细胞超微结构及增殖的影响。方法应用透射电镜观察AZT作用前后肿瘤细胞超微结构的变化,用MTT分析法对不同浓度AZT作用的卵巢癌HO-8910细胞进行药物敏感实验,并用流式细胞仪测定细胞周期的变化。结果 AZT作用后的HO-8910细胞生长明显受抑制,并有时间依赖关系和剂量依赖关系。形态学超微结构显示细胞器扩张和肿胀,核染色质边集,凝集成块。细胞周期中G1期细胞增多,S期细胞减少,并且可测到凋亡峰含量为14.2%。结论 AZT能明显抑制卵巢癌细胞的增殖,因而可为卵巢癌和其它肿瘤的治疗尝试一条新的有效的方法。
Objective To study the effect on ultrastructures and proliferation of ovarian cancer cells in vitro by reverse transcriptase inhibitors (AZT). Methods The ultrastructures of ovarian cancer cells were observed under transmission electron raicroscope.The drug sensitivity test was performed with MTT assay. The cell cycle was measured with fluocytometry (FCM). Results Proliferation of HO-8910 cells was significantly inhibited when treated with AZT, and the effect was time-dependent and dose-dependent. Under transmission electron microscope ,the chromatin concentrated near to the karyotheca. G1 phase ceEs increased and S phase cells decreased in cell cycle, the apoptotic peak was detected with FCM and reached 14.2%. Conclusion AZT can effectivelg inhibit the telomerase activity and the proliferation of tumor cells, and may provide an effective and novel approach for treating cancer.
