慢性髓性白血病cyclin E基因蛋白表达及临床意义

摘要 目的探讨CML患者Cyclin E基因蛋白表达特点。方法利用免疫组织化学实验方法检测34例CML患者Cyclin E基因蛋白。结果 CML患者Cyclin E基因蛋白表达明显高于对照组,有极显著性差异(P<0.001)。CML患者治疗前与治疗后Cyclin E阳性表达率也存在着明显差异(P<0.05),同时还观察了急变期/加速期、慢性期、缓解期Cyclin E基因蛋白表达,在急变期/加速期、慢性期时呈高表达状态与缓解期比较有显著性差异(P<0.05)。结论 Cyclin E基因蛋白表达与CML患者病情进展程度密切相关,该研究工作揭示CML的分子发病机制,同时对CML的诊断和疗效监测具有十分重要的临床应用价值。 Objctive To explore expression and significance of cyclin E protein in chronic myelogenous leukemia. Methods Immunohis-tochemistry was vised to analyze samples of 34 chronic myelogenous leukemias for their expression of cyclin E. Results Cyclin E was significantly higher expressed in CML than in normal samples (P < 0.001) . There was significant difference between untreated and treated patients (P < 0.05) . Cyclin E was higher expressed in accelerated phase/blast crisis phase and in chronic phase than complete remission (P < 0.05) . Conclusions Cyclin E gene expression is correlated with the clinical phase of CML, increased with disease progressed. The results showed that levels of cyclin E expression might be used as a the important direction marker for CML diagnosis and therapy.
