摘要
利用12个随机引物对来自云南省不同地理环境的立枯丝核菌RhizoctoniasolaniKühn第一融合群(AG-1)的13个菌株进行遗传分化关系的研究。结果表明受试菌株被标记的DNA谱带多态性检测率为100%,即受试菌株间几乎无同质的DNA谱带。利用UPGMA法构建分子系统树分析发现,遗传距离0.5将其划分为9个遗传聚类组,遗传距离0.86将其划分为6个遗传聚类组,而遗传距离0.95可将其划分为3个遗传聚类组。结果表明受试AG1融合群的13个菌株具明显的遗传分化。
Twelve RAPD primers were used to assess the genetic variation of 13 isolates of Rhizoctonia solani AG-1 from different localities of Yunnan province . A total of 182 polymorphic bands was amplified with 12 primers and no common bands were found among testd isolates. The result showed that the rate of polymorphism of the isolates was 100 percent. The dendrogram derived from RAPD data by Unweighted Pair Group Method using Arithmetic Average (UPGMA) suggested that 13 isolates could be divided into 9 subgroups with the genetic distance of 0.5, 6 subgroups with the genetic distance of 0.86, and 3 subgroups with the genetic distance of 0.95. It was proved that the isolates of AG-1 from different localities of Yunnan manifested abundant genetic diversity.
