白芷总香豆素类有效部位的纯化工艺研究 被引量:3

摘要 目的:研究白芷总香豆素类有效部位的萃取纯化工艺。方法:采用石油醚-乙酸乙酯联用法对白芷提取物进行纯化,以总香豆素及欧前胡素含量为考察指标,分别采用紫外分光光度法及高效液相色谱法进行含量测定,以确定最佳纯化工艺。结果:最佳纯化工艺为:白芷提取液2 g.m L-1,2倍量石油醚萃取1次,弃去,水层加2倍量乙酸乙酯萃取3次,合并乙酸乙酯层,总香豆素含量达50%以上,其中欧前胡素含量达4%以上。结论:纯化工艺稳定可行,可为白芷总香豆素制备提供参考。 Objective: To research the extraction and purification process of effective parts of Angelica dahurica total coumarin. Method: Petroleum ether and ethyl acetate were both used in the purification of Angelica dahurica. In order to choose the best purification methods, total coumarin and imperatorin were selected as indexes, and then the extration was determined by ultraviolet spectrophotometer and HPLC. Result: The best purification method was as following: Angelica dahurica extract 2g.m L-1, 2 times amounts of Petroleum ether extract one time, abandon petroleum ether, 2 times amounts of ethyl acetate extract 3 times, combine the ethyl acetate layer. The total coumarin content reached above 50%, in which imperatorin content was more than 4%.Conclusion: The extraction method is stable and feasible which can provide a reference for the preparation of Angelica dahurica total coumarin.
