摘要 目的 以含氯消毒剂对甲型流感病毒灭活试验效果的观察 ,探讨消毒剂杀灭病毒试验方法。方法 试验采用细胞感染法 ,以活细胞染色法 (MTT法 )和观察细胞病理变化 (CPE)法测定中和剂、消毒剂及中和产物对狗肾细胞 (MDCK)存活性的影响 ;通过流感病毒对细胞感染情况的观察、计算半数细胞感染剂量 (TCID50 )的值 ,确定消毒剂对病毒的杀灭效果。结果 含氯消毒剂有效氯含量 2 5 m g/L 时 ,作用于细胞 ,仍可影响细胞的存活性 ,以MTT法测定 ,细胞存活率仅为 2 6 .15 % ,与 CPE法测定含氯消毒剂对狗肾细胞存活性影响的结果基本一致 ;用MTT法筛选出的含 0 .0 7%硫代硫酸钠的 PBS的中和剂 ,可作为含氯消毒剂杀灭甲型流感病毒试验的中和剂 ;有效氯含量 4 0 0 mg/L、作用 10 m in,对甲型流感病毒的平均杀灭率为 99.91%。结论 以细胞感染法确定消毒剂对病毒杀灭效果的试验方法简便、可行。 OBJECTIVE To investigate a commercial chlorine releasing disinfectant, which is ratified by the Ministry of Public Health. Testing methods of disinfectant for killing virus were discussed through the observation of germicidal efficacy of this disinfectant to influenza virus.METHODS The method of virus infecting cells was adopted. Madin darby canine kidney (MDCK) was used as the sensitive cell. The cytopathic effect (CPE) and MTT assay were used to evaluate the effect of disinfectant, neutralizer and product of neutralization to the survival rate of cells. CPE caused by influenza virus, after which has been contacted with disinfectant, was used as the index to calculate the TCID 50 . The germicidal efficacy of disinfectant was calculated based on TCID 50 . RESULTS When the end available chlorine concentration of this chlorine releasing disinfectant is 25 mg/L, the survival rate of MDCK cell is only 26.15% by MTT method. This result is similar to the result by CPE method. The neutralizer for this chlorine releasing disinfectant is PBS with 0.07% Na 2S 2O 3 by MTT method. When the end available chlorine concentration is 400 mg/L with 10 minute contact time, the average killing rate is 99.91%.CONCLUSIONS The germicidal efficacy of disinfectant on influenza virus can be determined by the method of virus infecting cells.
