摘要
目的 :探讨氯沙坦 (Losartan)逆转自发性高血压大鼠 (SpontaneonslyHypertensiveRat ,SHR)左室重构与血管紧张素Ⅱ (AngiotensinⅡ ,AngⅡ )水平、内皮功能和细胞内钙代谢的关系。方法 :2 1只SHR随机分三组 :3月龄对照组 (SHR C3)、6月龄对照组 (SHR C6 )和Losartan治疗组 (SHR L) ,治疗组从 3月龄开始给予Losartan 2 0mg/kg·d-1治疗至 6月龄 ,治疗前后检测项目包含 :鼠尾收缩压 (SBP)、左室重量指数 (LVMI)、血浆和心肌组织AngⅡ水平、血清一氧化氮 (NO)、血浆内皮素 (ET)和淋巴细胞胞浆内游离钙 ([Ca2 + ]i)。结果 :治疗前与同龄正常血压对照大鼠 (WKY)比较 ,SHR的SBP、LVMI、血浆和心肌组织AngⅡ水平、ET水平、[Ca2 + ]i均显著性增加 ,而NO水平显著性下降 ,以上差别随鼠龄增长而加大 ;Losartan治疗后 ,SHR的SBP、LVMI、心肌组织AngⅡ水平、ET水平、[Ca2 + ]i均显著性下降 ,而NO水平显著性上升。结论 :Losartan逆转左室重构除与AT1受体阻滞有关外 ,可能还涉及降低组织AngⅡ水平、改善内皮功能和纠正细胞内钙代谢紊乱。
Objective:To investigate the relationship between the reversal effects of Losartan on the left ventricular remodeling and the effects of myocardiac Angiotensin Ⅱ (AngⅡ) level,endothelial function and intracellular calcium metabolism in SHR. Method:Twenty one spontaneously hypertensive rats (SHR) were randomly distributed to 3 groups:3 month control group (SHR C3),6 month control group (SHR C6) and Losartan group (SHR L). The 3 monthage SHRs from Losortan group were treated with Losortan(20 mg/kg·d -1 ) until the rats were 6 month by age.SBP,LVMI,plasma and myocardiac AngⅡ,serum Nitric Oxide(NO),plasma Endothelin(ET),peripheral lymphocyte cytosolic calcium([Ca 2+ ]i) were assessed before and after treatment. Results:Compared with WKYs,the SBP,LVMI,Plasma and myocardiac AngⅡ,ET,[Ca 2+ ]i were all significantly greater but NO was significantly less in SHRs.The difference between SHRs and WKYs became more prominent when they grew older. The SBP,LVMI,myocardiac AngⅡ,ET and [Ca 2+ ]i were all significantly decreased but NO was significantly increased in SHRs after Losartan therapy. Conclusions:Losartan can reverse the left ventricular remodeling not only by blocking AT 1 receptor but also by lowering tissue Ang Ⅱ,improving endothelial function and correcting the disorder of intracellular calcium metabolism.
