新疆黑果小檗根薄层定性鉴别及含量测定方法的建立

摘要 目的:建立黑果小檗根部化学成分的定性鉴别及含量测定方法。方法:采用薄层色谱法考察不同展开条件、展开剂比例、点样量、检视方式、薄层板、展开温度以确定最佳薄层色谱展开系统,并对黑果小檗根部化学成分进行定性鉴别及含量测定。结果:确定氯仿-乙酸乙酯-甲酸(4:4. 5:1. 5),薄层板为GF254硅胶薄层板,点样量为1μL,检测波长为254、366 nm,展开温度为25℃为最佳展开条件,黑果小檗根皮及根木质部中均含有盐酸巴马汀、盐酸小檗碱且盐酸巴马汀、盐酸盐酸小檗碱分别在0. 0211~3. 798、0. 675~4. 05范围内具有较好的线性关系(r=0. 9995、r=0. 9942),其中超声提取的根皮水提物中盐酸巴马汀含量最高,含量为2. 2920 mg/g;超声提取的根皮甲醇提取物中盐酸小檗碱含量最高,含量为3. 7640 mg/g。结论:本实验所建立的薄层展开方法稳定可行,含量测定方法操作简便、重现性好。 Objective:To establish the qualitative identification and content determination methods for chemical composition in Xinjiang Berberis heteropda Schrenk root.Methods:Methods:Thin-layer chromatography was used to determine the optimum thin-layer chromatographic system for different development conditions,the ratio of developer,spotting volume,inspection method,thin-layer plate and deployment temperature,and qualitative identification and content determination methods for chemical composition in Xingjiang Berberis heteropda Schrenk root.Results:Chloroform-ethyl acetate-formic acid(4∶4.5∶1.5)was determined,and the thin layer was a GF254 silica gel thin-layer plate.The sample amount was 1μL.The detection wavelength was254 nm and 366 nm.The development temperature was 25℃.Both the root bark and the root xylem of Xinjiang Berberis heteropda Schrenk contained palmatine hydrochloride,berberine hydrochloride and palmatine hydrochloride and berberine hydrochloride had a good linear relationship in the range of 0.0211-3.798 and 0.675-4.05(r=0.9995,r=0.9942),respectively.Among them,the content of palmatine hydrochloride in the water extract of ultrasonically extracted root bark was the highest and the content was 2.2920 mg/g.The content of berberine hydrochloride in the methanol extract of root barks extracted by ultrasonic was the highest and the content was 3.7640 mg/g.Conclusion:The thin layer development method established in this experiment is stable and feasible.The content determination method is easy to operate and has good reproducibility.
