流式细胞术在网织红细胞检测中的应用 被引量:3

摘要 目的评价流式细胞仪测定外周血中网织红细胞的性能。方法选取门诊体检及住院患者共300例,随机选择血标本,采用FACSCalibur型流式细胞仪进行网织红细胞计数,分析其重复性,并与传统煌焦油蓝染色显微镜法进行对比及做相关分析。结果流式细胞仪法与传统煌焦油蓝显微镜法关于网织红细胞低值、中值及高值计数分别比较,均差异无统计学意义(P>0.05);流式细胞仪的变异系数4.3%,传统煌焦油蓝显微镜法8.2%,两者比较差异有统计学意义(P<0.05),两种方法测定结果呈正相关关系(r=0.994,P<0.01)。结论流式细胞仪计数网织红细胞快速,结果准确,且重复性好,与传统煌焦油蓝染色显微镜法高度符合。 Objectives To evaluate the flow cytometric in reticulocyte detection of peripheral blood. Methods A total of 300 cases of outpatient and hospitalizd patients were selected in our hospital from March 2014 to July2014. Reticulocyte of random blood samples were detected by FACSCalibur flow cytometry. The repetition of FACSCalibur flow cytometry was analyzed and the correlation analyzed between FACSCalibur flow cytometry and traditional brilliant cresyl blue staining microscopy. Results There were all no significant differences about the reticulocyte counting of low,median and high value between FACSCalibur flow cytometry and traditional brilliant cresyl blue staining microscopy respectively. The coefficient of variation of flow cytometry was 4. 3% and that the traditional brilliant cresyl blue staining microscopy was 8. 2%. There was significant difference between two methods( P < 0. 05). The correlation between the determination results of FACSCalibur flow cytometry and traditional brilliant cresyl blue staining microscopy was positive( r = 0. 994,P < 0. 01). Conclusion Reticulocyte counting by Flow cytometric is fast,accurate,and good reproducibility. The results of reticulocyte counting by Flow cytometric are highly consistent with the traditional brilliant cresyl blue staining microscopy.
