彗星试验检测武汉市氯化饮水有机提取物对HepG2 DNA的损伤 被引量:16

摘要 目的 研究武汉市主要水源 C江、D湖、H水的氯化饮水有机提取物对 Hep G2 DNA的损伤。方法 氯化饮水有机提取物染毒 Hep G2后 ,用单细胞凝胶电泳 (亦称彗星试验 )检测 DNA损伤。结果 氯化饮水有机提取物诱导DNA损伤的最低浓度 ,C江、D湖、H水丰水期 (8月份 )均为 1.6 7m l/ ml培养液 ,平水期 (3月份 )分别为 16 7、 16 .7、1.6 7m l/ ml培养液 ;最高浓度 (16 7m l/ ml培养液 )氯化饮水有机提取物诱导的 Hep G2 DNA迁移度 ,C江、D湖的丰水期均明显高于平水期 (均 P<0 .0 5 ) ;最高浓度氯化饮水有机提取物诱导 Hep G2 DNA损伤的强度 ,H水 >D湖和 C江 (均 P<0 .0 1)。结论 武汉市主要水源 C江、D湖、H水的氯化饮水有机提取物对 Hep G2 DNA有损伤作用 ,损伤作用的严重程度 H水 >D湖 >C江 ,丰水期 > Objective To study DNA damage in HepG2 cells exposed to organic extracts from chlorinated drinking water (CDW) from three raw water sources of C river, H river and D lake in Wuhan. Methods DNA damage was tested in HepG2 cells exposed to organic extracts from CDW by using single cell gel electrophoresis assay (SCGE). Results The lowest concentrations inducing DNA damage were 1 67 ml/ml culture in organic extracts from CDW sampled on August and 167, 16 7 and 1 67 ml/ml culture in organic extracts of CDW sampled on March from raw water sources of C river, D lake and H river respectively. At the highest concentration (167 ml/ml culture), DNA migration was increased in organic extracts of CDW sampled from raw water sources of C river and D lake on August as compared with those on March( P <0 05), and DNA breakage was stronger in organic extracts of CDW sampled from raw water source of H river than that of C river and D lake. Conclusion Organic extracts of CDW from three raw water sources of C river, H river and D lake in Wuhan could induce DNA strand breaks in HepG2 cells. DNA damage levels induced by organic extracts of CDW sampled on August were higher than on March. DNA damage levels by organic extracts of CDW from raw water source of H river were the highest of three raw water sources.
