摘要
目的 :研究槐果碱对感染CVB3 小鼠NK细胞和IL - 2活性的影响。方法 :病毒接种法制备CVB3 病毒性心肌炎小鼠模型 ,用MTT比色分析法与ELISA法分别检测小鼠脾脏中NK细胞活性与IL - 2的诱生量。结果 :病毒模型组NK活性明显升高 ,IL - 2活性降低。槐果碱 5 0mg .kg-1.d-1和 2 5mg .kg-1.d-1两个剂量ig可明显升高正常小鼠NK活性。但槐果碱各治疗剂量对CVB3 心肌炎小鼠NK细胞活性无明显影响。槐果碱 5 0mg .kg-1.d-1ig治疗组可明显增加脾脏IL - 2诱生量。结论 :槐果碱升高CVB3 感染小鼠的IL - 2活性 ,这可能是其抗CVB3 免疫机理之一。
Objective: To study the effect of Sophocarpine on activities of NK and IL-2 in murine model with coxsackie virus B_3-induced viral myocarditis. Methods: the activities of splenic NK were measured by MTT assay and the content of splenic IL-2 was determined by ELISA method respectively, all data of experimental groups and blank control group were collected and compared statistically. Results: The activity of NK in murine model with coxsackie virus B_3-induced myocarditis was significantly increased while the content of splenic IL-2 decreased, compared with those in healthy control group. The activities of splenic NK in healthy group treated with sophocarpine 50 mg. kg -1 .d -1 ig and 25 mg.kg -1 .d -1 ig were significantly increased in contrast to those in healthy control group received normal saline only. No significant difference was found in the activity of NK between the coxsackie B_3-induced myocarditis mouse model treated with sophocarpine and the blank control group. The content of splenic IL-2 in mouse model treated with sophocarpine 50mg.kg -1 .d -1 ig was significantly increased, compared with that in the blank control group. Conclusions: The activity of IL-2 increases significantly in mouse model treated with sophocarpine, as compared with blank control group.
