摘要
以树莓芽作为外植体,在MS、1/2MS、WPM、Anderson4种基本培养基中培养40d后,芽诱导率分别为90.48%、64.4%、61.7%、67.5%。通过单因子试验和正交试验进行细胞分裂素和生长素的筛选,选择出IBA和6 BA2种激素,其最佳组合是IBA0.1mg/L+6 BA1.0mg/L,40d后芽增殖倍数为3.7。1/2MS作为生根培养基,附加IBA0.1mg/L时,根诱导率为80.0%,平均根长为2.2cm。树莓的组织培养中,蔗糖浓度最宜为30g/L。
Buds of Rubus idaeus is often used as the material for tissue culture in order to improve the productive rate. It is found in this experiment that the most suitable cultural media for bud induction is MS+IBA0.1 mg/L+6BA1.0 mg/L, in which the inductivity can amount to 82% after 40 day's cultivation. The best rootinducing cultural medium is 1/2MS+IBA0.1 mg/L, in which 80% of the material can produce their roots after 40 day's cultivation and the root can grow as long as 2.2 cm. The best sucrose concentration is 30 g/L.
