羟基磷灰石纳米粒子对人肝癌细胞增殖及细胞周期的作用研究(英文) 被引量:10

摘要 目的 :研究羟基磷灰石 (HAP)纳米粒子体外抗肝癌作用的机理。方法 :采用形态学观察和MTT法检测HAP纳米粒子作用于人肝癌细胞系Bel 74 0 2的量效和时效关系。流式细胞仪分析细胞周期的时相变化。结果 :HAP纳米粒子在体外对人肝癌细胞系Bel 74 0 2具有明显的抑制作用 ,并呈现良好的量效和时效关系。肝癌细胞的生长阻滞于G1期。结论 :HAP纳米粒子具有体外抗肝癌作用 ,细胞增殖阻滞于G1期 ,阻断细胞周期的进展 ,导致癌细胞胀亡。 objective To explore the mechanism in which hepatocellular carcinoma cells were inhibited by HAP nanopaticles in vitro.Methods The time and dose effect dependent manner were investigated by the MTT method and morphology observation.The changes of cycle phase were analyzed by FCM.Results There was a good time and dose effect dependent manner and Bel 7402 hepatocellular carcinoma cells were inhibited by HAP nanopaticles.The proliferation of the human hepatocellular carcinoma cells was arrested at G1 phase of cell cycles.Conclusions HAP nanopaticles have the effect of anti hepatocellular carcinoma in vitro.The cell proliferation was arrested at G1 phase and progress of the cell cycle was blocked.Thus the tumor cells oncosis.
