摘要
目的 :通过对创伤弧菌 (VV)的培养研究 ,选择最佳优化培养条件 ,提高该菌的检出率。方法 :用酶标仪测定各生长条件下的细菌吸光度值。结果 :VV在各测试条件中 ,含 1%NaCl培养 10h细菌吸光度达峰值 ;6 4 0mg/L血红蛋白于 6h达峰值 ;10 %蛋白胨于 8h达峰值 ;在pH值为 8.0~ 8.5、温度为 37℃时 ,细菌吸光度达峰值。结论 :VV最佳优化培养条件为 :1%NaCl、6 4 0mg/L血红蛋白、10 %蛋白胨、pH 8.0~ 8.5、温度 37℃。
Objective:To study optimum culture condition and improve the detection of vibrio vulnificus(VV) by culturing VV.Methods:Determine the VV's absorbance value of different condition with automatic microplate.Results:the absorbance value reached the peak in 10 hour,6 hour and 8 hour,respectively, when the basic media contain 1% NaCl,640 mg/L Hb, or 10% peptone. Absorbance value also reached the highest when pH value was 8.0 to 8.5 and the temperature was 37 ℃.Conclusion:The optimum culture condition to VV were the basic media containing 1% NaCl,640 mg/L Hb, 10% peptone,pH value from 8.0 to 8.5 and temperature was 37 ℃,which can be used to study the fast detection of VV.
