摘要 目的 :观察低密度脂蛋白 ( ox L DL)对培养血管平滑肌细胞 ( VSMC)凋亡的影响 ,探讨 ox L DL 致VSMC凋亡的相关机制。方法 :体外培养 Wistar大鼠 VSMC并确认细胞指数生长期 ,用苔盼蓝拒染计数方法观察 ox L DL刺激 VSMC对增殖的影响 ,流式细胞分析检测 ox L DL刺激 VSMC凋亡以及 COX 2抑制剂NS 398对凋亡的影响。结果 :体外培养的 VSMC正常生长后 ,用 35 mg/ L和 5 0 mg/ L ox L DL可使培养VSMC的生长期提前到 12~ 2 4h,表现出明显细胞增殖 ;NS 398干预 ox L DL 组 VSMC增殖受到减弱。流式细胞分析 ox L DL 组 VSMC早期凋亡率较对照组明显增加 ,而 NS 398、ox L DL 刺激 VSMC凋亡阳性率亦较对照组明显增加。结论 :ox L DL 刺激可使培养 VSMC表现明显细胞增殖 ,虽 NS 398可减弱 VSMC的增殖 ,但却协同 ox L DL 导致凋亡作用的增强。 Objective:To investigate the apoptosis in vascular smooth muscle cell (VSMC) induced by oxidated lowdensity lipoprotein cholesterol (oxLDL),and to discuss the relative mechanism of the apoptosis of VSMC.Methods:The growth index of VSMC of Wistar rat was determined.By using trypan blue to count unstaining cells,the proliferative effect of oxLDL on culturing VSMC was observed.With flow cytometer,the apoptosis of VSMC induced by oxLDL and the inhibitory effect of cyclooxygenase2 (COX2) inhibitor NS398 on apoptosis were determined.Results:After VSMC grew normally in culture, oxLDL in concentrations of 35 mg/L and 50 mg/L could advance the growth period of VSMC to between 12 and 24 hours,and the proliferation of VSMC were obvious.The proliferation of VSMC induced by oxLDL were inhibited by NS398.The rate of apoptosis of VSMC in oxLDL and NS398 groups were higher than that of normal control.Conclusion:oxLDL could induce an obvious proliferation of VSMC in culture. Although NS398 can decrease the proliferation of VSMC,it could enhance oxLDL to induce apoptosis.
