摘要
目的 :探讨红藻氨酸 (KA)致痈大鼠海马神经细胞凋亡与野生型p5 3(wtp5 3)蛋白表达的关系。方法 :采用KA诱导癫痫发作大鼠模型。以TUNEL方法标记DNA片段 ,原位检测凋亡细胞 ;免疫组化SP法检测wtp5 3蛋白。结果 :注射KA 72h后 ,癫痫发作大鼠海马CA1区凋亡神经细胞达高峰及wtp5 3蛋白表达在 2 4h大量出现 ;wtp5 3蛋白表达与细胞凋亡呈正相关。结论 :癫痫神经细胞凋亡与wtp5 3表达密切相关 ;痫性发作使wtp5 3表达上调 ,从而诱导神经细胞凋亡。
Objective: To explore the relationship between neuronal apoptosis and wild-type p53(wtp53) protein expression in the hippoeampus of the rata with kainic-acid-induced seizures.Methods:With the model of kainic-acid-induced epilepsy used, apoptotic cell death was identified with the terminal deoxynucleotidyl transferase-mediated dUTP-biotin in situ nick end labeling(TUNEL) technique to detect DNA fragmentation and wtp53 protion exprression was determined with immunohistochemisty(SP) .Results: After kainic acid injections for 72 hours, significant increase in the apoptotic neurons andmany wtp53-positive neurons were observed at the 24th in the rats with seizr, res. Correlation analysis showed that neuronal apoptosis had positive correlation with wtp53 protein exprssion,Conclusions: The results suggest that wtp53 protein expression has a close relation to neuronal apoptosis after seizures, the seizures make wtp53 protein expression increase, thus induce neuronal apoptosis.
