摘要
选择毕赤酵母GS115作为表达宿主,对抗菌肽Spinigerinα的突变体Spinigerinα′的基因进行胞外分泌表达,并选用致病性大肠杆菌、致病性的肠炎沙门氏菌、金黄色葡萄球菌作为敏感菌株,采用琼脂空穴扩散法对表达液进行抑菌效果的研究,并将其抑菌效果与突变前的抗菌肽Spinigerinα进行对比。结果表明,抗菌肽Spinigerinα′-1的抑菌活性降低,Spinigerinα′-2抑菌活性稍有增加,Spinigerinα′-3的抑菌活性最强,而Spinigerinα′-4和5酵母表达的蛋白无抑菌活性。为抗菌肽抑菌效果提高的研究奠定重要的基础。
In this study, Pichia pastoris GS115 as expression host is for antimicrobial peptides Spinigerinα' gene extracellular secretion expression. Using agar hole diffusion method, on E.coli-K12D31, Staphylococcus aureus and Salmonella enteritidis as sensitive strains, for antimicrobial peptides Spinigerin α' expression liquid antibacterial. The results show that the antibacterial activity of the Spinigerinα'-1 reduces, Spinigerinα'-2 antibacterial activity increases slightly, Spinigerinα'-3 inhibition activity is strongest, and Spinigerin α'-4 and the 5 yeast expression protein have no antibacterial activity. The trial has laid an important foundation for research of antimicrobial peptides antibacterial effect improved.
