摘要
目的 :观察天保宁对阿霉素所致大鼠心肌损伤时心肌肌球蛋白重链的变化及心肌结构的改变。方法 :采用雄性SD大鼠 30只 ,随机分为 3组 :(1)正常对照组 :ip等体积生理盐水 ;(2 )阿霉素组 (Doxorbincin ,Adrinmycin ,ADR) :于实验开始后第 2、4天ipADR 1mg kg ;第 6、8天ipADR2mg/kg ;第 10、12天ipADR 3mg kg ;第 14、16天ipADR 4mg/kg ,16d累计用药剂量达 2 0mg kg。 (3)天保宁组 (ADR +天保宁 ) :ADR的剂量用法同ADR单用组 ,天保宁 (浙江康恩贝制药公司 ) 4 5 0mg/kg隔日灌服共 8d停药 2 4h后 ,称体重、心肌重量、左心室重量及心肌蛋白的提取、肌球蛋白重链(MHC)分析 ,电镜观察心肌细胞超微结构的变化。结果 :天保宁组大鼠的体重、心体比及左室心体比 (LVM BW )均较ADR组有显著改善 (vsP <0 0 1)。电镜下可见 ,ADR组心肌细胞严重损伤 ,提示有心衰发生 ,天保宁组大致正常。同时 ,天保宁组中α MHC向 β MHC转变减轻 ,α / β MHC较ADR组低 (P <0 0 5 )。结论 :天保宁可改善ADR对心肌损伤时的毒性反应 ,特别是减轻ADR对心肌肌球蛋白重链中α MHC向β MHC转变 ,具有抗重塑和抗心衰的作用。
齌o study the effects of Tian Bao Ning on Mysion Heavy Chain (MHC) and myocardial structure in rats heart injury induced by doxorbincin (ADR). Methods: 30 males SD rats were divided into: (1) control group ( n =10); (2)ADR group ( n =10 20mg/kg ADR given intermittently with in 16 days);(3)Tian Bao Ning & ADR( n =10 480mg/kg Tian Bao Ning given intermittently with in 16 days and ADR given as ADR group). Body weight, left ventricle mass to body weight ratio(LVM/bw), α MHC and β MHC were measured. The ultrastructural changes of cardiomyocyles were examed electromicrographically. Results: Shifts of myosin profiles from α MHC to β MHC occurred in ADR group with a decrease in α MHC(vs control P <0 01) and the increase in β MHC(vs control P <0 01). Ultrastructural changes of cardiomyocytes in rats heart failure were electromicroscopically observed in ADR group. After the treatment of Tian Bao Ning. The myosin profiles from β MHC to α MHC, namely, an increase in α MHC (Vs ADR group P <0 01) and a decrease in β MHC (Vs ADR Group P <0 01) were observed. There is significant decrease in LVM/BW(Vs ADR Group P <0 01). Conclusion: Tian Bao Ning may alleviate the pathological changes in cardiaomyocyles caused by ADR and reverse the changes in the expression of Myosin Heavy Chain in myocardiac cells.
