利用叶绿体DNAtrnL-F序列探讨石蒜属种间关系 被引量:1

INVESTIGATION ON THE INTERSPECIFIC RELATIONSHIPS OF Lycoris(AMARYLLIDACEAE) BASED ON TRNL-F SEQUENCES

摘要 本文利用叶绿体DNA trnL-F区序列,并结合现有的形态和染色体核型资料,分析了石蒜属15个种及2个人工杂种的种间关系,结果表明:在系统发育树上,石蒜属构成一个单系类群,供试材料组成两个主要分支,系统树上亲缘关系近的类群聚在一起构成次级分支,较好揭示了各种的种间关系。trnL-F序列揭示的石蒜属种间关系与染色体分类的结果部分一致,主要表现在具有近端部着丝粒染色体的种与具有中部和端部着丝粒染色体的种各成一支,这与形态和染色体分类结果一致;不同之处在于具有中部、端部和近端部着丝粒染色体的种分散在其它两支内,这证明了具有三种类型染色体组的石蒜是杂交起源的假设。结合2个人工杂种分析,发现短蕊石蒜的近端部着丝粒染色体来源于换锦花;麦秆石蒜近端部着丝粒染色体来源于矮小石蒜。 In this paper,the trnL-F region of chloroplast DNA(cpDNA) was sequenced from selected species for interspecific relationships analysis.The species based on cpDNA trnL-F region was clearly classified into two main groups which included several subclades.On the phylogenetic tree,closely related species grouped together in the clade showing more precise and resolved relationships among studied taxa,the result of trnL-F sequences analyses is concordant with morphological and karyotype classification in certain respects and discordant in others: the concordancy shows all the subtelocentric chromosome species formed the first clade,and all the telocentric and metacentric chromosomes species formed the second clade.The discordancy indicates all with mixtures of subtelocentric,telocentric and metacentric chromosomes species dispersed in the two clades which reinforced the hypothesis of hybrid origin of these species.Comparing two artificial hybrids,trnL-F sequence analysis was also confirmed that the subtelocentric chromosome of L.caldwellii derived from L.sprengeri,while L.straminea was from L.radiata var.pumila.
