摘要
目的 :建立链脲佐菌素 ( streptozocin,STZ)糖尿病大鼠视网膜病理模型。方法 :建立糖尿病大鼠模型 ,随机分为正常对照组 ( M)、糖尿病 1个月组 ( M1)、3个月组 ( M3 )和 5个月组 ( M5)。分别观察视网膜铺片及石蜡切片变化。结果 :普通石蜡切片中 M、M1未见异常改变 ,从 M3 开始出现异常改变 ,以 M5最明显。表现为视网膜水肿 ,尤其内核层。细胞排列紊乱 ,内核层血管周围水肿更明显 ,节细胞数量减少 ,静脉扩张、渗出 ,并有出血。视网膜消化铺片 :1周细胞数量 :糖尿病 M1组与 M组相比差异无显著性 ( P>0 .0 5 ) ,而 M3 组及 M5组与 M组相比明显减少 ( P<0 .0 1 ,P<0 .0 0 1 )。 2毛细血管形态 :以 M5组时病变最重 ,可见毛细血管栓塞、无细胞毛细血管及毛细血管无灌注。结论
Objective:To evaluate paraffin slide, retinal digest preparation and ultrastrucure of diabetic retinopathy rat in dciced by STZ. Methods:Diabetes model of rat was induced in rats with a single injection of streptozotocin (STZ) given intraperitoneally .Aminals were randomly divided into control group (M), diabetic for 1 month group (M 1), diabetic for 3 months group (M 3) and 5 months group (M 5), respectively. To observe the morphology in retinal paraffin slide and retinal digest preparation and ultrastrucure of retina by transparent electron microscopy. Results:No change was found in M and M 1 on paraffin slide. Different degree of lesion was found in M 3 on paraffin slide. The most dramatic lesion was observed in M 5. It showed as following:The retinal swell, the most seriously swell was in membrana granulosa interna. Disorder of the morphology of cell. The membrana granulosa interna swell obviously around vas. The number of ganglionices decreased. The retinal venae expand, exude and hemorrhage. Retinal digest preparation: ①Number of pericytes: There was no difference M 1 and M ( P >0.05); The number of pericytes decreased obviously in M 3 and M 5 ,there was no differencecompared with M ( P <0.01, P <0.001). ②Morphology of capillary: The most seriously change was seen in M 5, including thrombus of capillary, lack of capillary, and lost irrigation of capillary. Conclusion:Diabetic retinopathy induced by STZ in rat may represent background diabetic retinopathy in human being.
