摘要
目的 观察慢性束缚应激模型大鼠相关脑区 (下丘脑、垂体、海马、皮层 )前阿黑皮素 (POMC)mRNA含量变化以及中药逍遥散的影响。方法 用反转录 聚合酶链反应 (RT PCR)和图像分析方法测定相关脑区POMCmRNA含量变化。结果 2 1d模型组较正常对照组POMC 1基因表达在四个脑区都显著下调且差异有统计学意义 ,疏肝药组较 2 1d模型组POMC 1基因表达在下丘脑和皮层显著下调 (P <0 0 1 ) ,在海马上调且差异有显著性 (P <0 0 5)。 2 1d模型组较正常对照组POMC 2基因表达在皮层区上调且差异有统计学意义 ,疏肝药组较 2 1d模型组POMC 2基因表达在皮层下调 (P <0 0 5)。结论 疏肝药组对慢性束缚应激中枢神经肽POMC的调节靶点在下丘脑、垂体、海马和皮层 ,充分证实疏肝药组对于POMC的调节靶点在下丘脑、边缘系统及皮层中枢等 ,推测疏肝药组同时有多靶点、双向调节 。
Objective To observe the amount change of proopiomelanocortin mRNA (POMC mRNA) of relative brain zones (hypothalamus,pituitary,hippocampus,cortex) and effect of xiaoyaosan (ease powder) on the chronic restrained stress model rats.Methods RT PCR and graphic analysis methods were applied to test the amount change of POMC mRNA of relative brain zones.Results The model group gene expression of POMC 1 was markedly down modulated in four brain zones,the expression of POMC 2 was up modulated in cortex zone and the difference was significant between the model group and the control group ( P <0 05).In the group of soothing the liver medicine,the gene expression of POMC 2 was down modulated in cortex zone and gene expression of POMC 1 was down modulated in hypothalamus and cortex zone, the difference was significant between it and the model group ( P <0 01) and up modulated in hippocampus and the difference is significant ( P <0 05).Conclusion The modulating points of soothing the liver group on central neuropeptide of chronic restrained stress model are respectively in hypothalamus,pituitary,hippocampus and cortex.The modulating targets are in hypothalamus,the limbic system and the cortex center.It can be suggested that medicines of soothing the liver simultaneously have poly target and dual modulating action and be involved in the integration function of nerve endocrine immune net.
