摘要
目的 :了解单纯疱疹病毒性脑炎 (HSE)小鼠脑组织内的ICAM 1mRNA动态变化及药物的影响。方法 :采用RT PCR方法半定量检测小鼠HSE治疗前后ICAM 1mRNA的表达 ,并给予阿昔洛韦 (ACV)及地塞米松 (DEX)治疗 ,用透射电镜观察药物治疗后脑细胞结构的变化。结果 :HSE小鼠在感染后第 3天ICAM 1mRNA的表达开始增加 ,第 4天达高峰 ,第 5天后逐渐下降 ;用药物干预的小鼠脑神经细胞改变较轻微 ,未找到病毒颗粒 ,毛细血管周围水肿减轻。结论 :ICAM 1mRNA是HSE炎症反应发生过程中的重要环节之一 ,HSE时早期给予ACV +DEX治疗对HSE脑细胞结构有明显保护作用。
Objective:To study the dynamic changes of the brain cell ICAM 1mRNA and the effection of the drugs in the mice with herpes simplex virus encephalitis.Methods:Using PT PCR half quantitative determination to detect the expression of ICAM 1mRNA in the mice of HSE that anterior and posterior the treatment.Treated with ACV+DEX,using transmission electron microscopy,the changes of the structures of the HSE mice brain cell is observed.Results:The ICAM 1mRNA expression of the mice with HSE begins to increase at the third day,attains the peak at the forth day,and descends gradually after the fifth day.Treated with ACV+DEX,the structure of HSE brain cell did not change too much and the virion was not found under transmission electron microscopy,the edema of the brain vessels of the HSE mice was attenuated.Conclusion:ICAM 1mRNA is one of the important links during the HSE inflammatory reaction.In early phase the administration of ACV+DEX,there is significant protection of the brain cells in the HSE.
